Summary: Ferroptosis, a newly discovered form of programmed cell death, is characterized by peroxidative damage to polyunsaturated-fatty-acid-containing phospholipids and is highly dependent on iron ions. It is closely associated with the pathogenesis of various diseases and is regulated by multiple signaling pathways, including the MAPK-AMPK pathway. This review summarizes the molecular mechanisms underlying the regulation of ferroptosis by the MAPK-AMPK pathway and discusses the potential therapeutic strategies targeting this pathway.

Summary: In this study, we found that overexpression of SLC40A1 can inhibit the proliferation, migration, and invasion of hepatocellular carcinoma (HCC) cells by activating the autophagy pathway. SLC40A1 regulates the autophagy level of MHCC-97H through the AMPK/mTOR/ULK1 and AMPK/ULK1 signaling pathways.

Ebp1 and its downstream signaling pathways may be potential therapeutic targets for HCC.

Summary: This study explores the synergistic anticancer activities of olaparib combined with a p38 MAPK inhibitor, SB203580, on non-small cell lung carcinoma A549 cells. The results demonstrate that while olaparib and SB203580 individually reduce cell viability in a dose-dependent manner, their combination synergistically reduces cell viability. The combination treatment enhances the anticancer potential of olaparib by increasing DNA damage-induced apoptosis and inhibiting telomerase activity.

Summary: The study aimed to investigate the cytotoxic effects of the AMPK inhibitor Compound C in chronic myeloid leukemia (CML) cell lines. Compound C induced apoptosis in certain CML cell lines, and its effects were enhanced by the silencing of SIRT3. Autophagy and downregulation of MCL1 were observed in response to Compound C treatment. The study suggests that AMPK inhibition is a promising strategy for improving CML therapy.

Summary: This study demonstrates that aberrantly activated CCRK signaling in chronic liver diseases can reprogram the immunosuppressive microenvironment to promote cancer metastasis to the liver. Overexpression of CCRK increases the metastasis of melanoma and colorectal cancer to the liver, while depletion of these signals reduces the risk of metastasis.

Summary: Liver resection is an established treatment for colorectal liver metastasis (CLM), but its role in non-colorectal liver metastasis (NCLM) is less clear. A study compared outcomes of curative hepatectomy for NCLM and CLM, finding no significant difference in 5-year overall and recurrence-free survival rates. Pre-operative systemic therapy, multifocal tumors, tumor size, R1 resection, and severe complications were identified as prognostic factors for poor overall survival.

Summary: Obesity and hypercholesterolemia are major risk factors for hepatocellular carcinoma. This study found that obesity leads to hepatic cholesterol accumulation, which suppresses natural killer T (NKT) cell-mediated antitumor immunosurveillance. By lowering cholesterol and inhibiting cholesterol biosynthesis, the function of NKT cells can be restored to prevent HCC development promoted by obesity.

Independent prognostic factors included age, ASA status, preoperative albumin, tumor number, and presence of microvascular invasion. The nomogram showed high accuracy in predicting long-term survival outcomes.
