Gracilibacillus xinjiangensis sp nov., a new member of the genus Gracilibacillus isolated from Xinjiang region, China

A Gram-positive, endospore-forming, rod-shaped bacterium, designated isolate J2(T) was isolated from a soil sample from Xinjiang Uyghur Autonomous Region, China. The isolate was observed to grow at 16-46 A degrees C and pH 6.5-8.0. Chemotaxonomic analysis showed menaquinone-7 (MK-7) to be the major isoprenoid quinone; diphosphatidylglycerol, phosphatidylglycerol, one aminophospholipid, two phosphoglycolipids and one glycolipid as the major cellular polar lipids; and anteiso-C-15:0, iso-C-15:0, anteiso-C-17:0 and C-16:0 as the major fatty acids. Comparative analyses of the 16S rRNA gene sequence showed that strain J2(T) is most closely related to Gracilibacillus ureilyticus (with 98.8 % similarity), Gracilibacillus dipsosauri (97.2 %), Gracilibacillus quinghaiensis (97.1 %) and Gracilibacillus thailandensis (97.0 %). The DNA-DNA reassociation values between strain J2(T) and G. ureilyticus MF38(T), G. dipsosauri DD1(T), G. quinghaiensis YIM-C229(T) and G. thailandensis TP2-8(T) were 29.8 +/- A 3.7, 23.0 +/- A 3.5, 15.8 +/- A 4.9 and 15.9 +/- A 5.0 %, respectively. The genomic DNA G+C content of strain J2(T) was determined to be 36.5 mol%. Based on these data, strain J2(T) is considered as a novel species of the genus Gracilibacillus, for which the name Gracilibacillus xinjiangensis sp. nov. is proposed. The type species is J2(T) (= CGMCC 1.12449(T) = JCM 18859(T)).

The Role of Glycoside Hydrolases in S. gordonii and C. albicans Interactions

Summary: This study revealed the glycoside hydrolase-related cross-kingdom communications between Streptococcus gordonii and Candida albicans under the low-fermentable-carbohydrate condition. We demonstrated that Candida albicans can promote the growth and metabolic activities of Streptococcus gordonii by elevating the activities of cell-wall-anchored glycoside hydrolases of Streptococcus gordonii. The Candida albicans gene TEC1 is critical for this cross-kingdom metabolic communication.

The interactions between oral-gut axis microbiota and Helicobacter pylori

Summary: In the human body, different microbial habitats exhibit distinct microbial population patterns and play a crucial role in the development of diseases. The oral cavity and gut are complex and interdependent microbial habitats, with Helicobacter pylori being one of the most important pathogens from the digestive tract. H. pylori infections can disrupt the normal gastric environment and enhance microbial interactions between the oral cavity and gut, thus reshaping the oral and gut microbiomes. The interaction between the oral-gut axis microbiota and H. pylori plays a significant role in H. pylori's colonization, infection, and pathogenicity. The balance of the microecology of the oral-gut axis can be altered by both H. pylori infection and eradication, which can impact the occurrence and progress of related diseases. The shift of oral-gut axis microbiota and their interactions with H. pylori may serve as potential targets for the diagnosis and treatment of H. pylori infections.

Candida albicans CHK1 gene regulates its cross-kingdom interactions with Streptococcus mutans to promote caries

Summary: This study demonstrated that Candida albicans CHK1 gene plays a crucial role in the development of dental caries by promoting the growth, biofilm formation, and exopolysaccharides production of Streptococcus mutans. The upregulation of S. mutans vicR and vicK genes by C. albicans CHK1 gene enhances the expression of EPS biosynthesis genes, leading to increased EPS production and biofilm formation. In a rat caries model, coinfection with chk1 Delta/Delta and S. mutans resulted in decreased colonization of S. mutans and less severe caries compared to other combinations.

The Preventive Effect of A Magnetic Nanoparticle-Modified Root Canal Sealer on Persistent Apical Periodontitis

Summary: Persistent apical periodontitis is a critical challenge in root canal therapy. Developing root canal filling materials with continuous antibacterial effects and tight seal is important to prevent treatment failure and persistent apical periodontitis. In this study, we modified the EndoREZ root canal sealer by adding the antibacterial material DMADDM and magnetic nanoparticles (MNPs). The modified sealer showed good mechanical properties, biocompatibility, and enhanced antibacterial effects on multispecies biofilms under a magnetic field. In vivo study demonstrated its inhibitory effect on persistent apical periodontitis with minimal resorption and slight increase in periodontal ligament space.

The Bio-Aging of Biofilms on Behalf of Various Oral Status on Different Titanium Implant Materials

Summary: This study investigated the effects of bio-aging on titanium implants in the oral microenvironment. Different surface treatments were used, and biofilms were established using different bacteria and saliva from healthy individuals and patients with periodontitis. After 30 days of bio-aging, the surface morphology, chemical composition, and water contact angles were measured. The adhesion of cells and biofilms was also evaluated. The results showed that the polished samples had no significant changes, while the MAO- and SLA-treated samples showed mild corrosion. The HA-coated samples were the most vulnerable, especially when exposed to saliva from patients with periodontitis. These findings highlight the importance of oral hygiene and periodontal health for long-term implant success.

Application of Fluorescence In Situ Hybridization (FISH) in Oral Microbial Detection

Summary: There are various microorganisms in the oral cavity that contribute to the occurrence and development of oral diseases. Fluorescence in situ hybridization (FISH) is a powerful tool for identifying and locating oral microorganisms, with advantages of rapidity, safety, high sensitivity, and specificity. FISH enables the identification and quantification of different oral microorganisms simultaneously, as well as visualization and distribution analysis of microbial communities in the oral biofilm.

Correspondence on Structural Insight into the Catalytic Mechanism of the Endoperoxide Synthase FtmOx1

Summary: In their recent publication, Cen, Wang, Zhou et al. reported the crystal structure of a ternary complex of the non-heme iron endoperoxidase FtmOx1 and concluded that it supports the CarC-like mechanistic model for catalysis. However, they failed to accurately describe the CarC-like and COX-like models and address their differences, and their data is consistent with the COX-like model after careful analysis.
