Summary: This study investigated the pharmacokinetic/pharmacodynamic relationships of linezolid in the treatment of tuberculosis, showing that time above minimum inhibitory concentration (T->MIC) was most correlated with efficacy, but area under the concentration-time curve over MIC (AUC/MIC) was equally predictive in growth-constrained conditions. Additionally, red blood cell counts were strongly correlated with LW minimum concentration (C-min).

Summary: Members of the genus Corynebacterium are increasingly recognized as pathobionts and can be very resistant to antimicrobial agents. A multicenter study revealed that multiple Corynebacterium species can rapidly develop daptomycin nonsusceptibility, including high-level daptomycin resistance, after a short exposure period. Differences in daptomycin MIC values between brands of medium were observed in a significant percentage of isolates.

Summary: Improper use of antibiotics can lead to poor treatment effectiveness and increased bacterial resistance. In this study, ultrafiltration probes were placed in the ileum of piglets with the help of anesthesia. The concentration of DAN in the ileum ultrafiltrate was significantly higher than in plasma, and PK parameters for both compartments were calculated using WinNonlin software. The study also found that the killing mechanism of DAN against E. coli is concentration-dependent, providing insights for rational dosage schedule design.

Summary: This study explored the restriction of Staphylococcus aureus resistance to linezolid/daptomycin combinations, showing that the combination therapy prevented the enrichment of linezolid-resistant mutants and restricted the daptomycin-resistant mutants. The anti-mutant effects of the combinations were attributed to the lengthening of time above MPC of both antibiotics, which suggests that T->MPC is a reliable predictor of the efficacy of antibiotic combinations against resistant mutants in in vitro dynamic models.

Summary: The study aimed to determine the efficacy of gepotidacin against E. coli and its relationship with pharmacokinetic-pharmacodynamic (PK-PD) indexes, as well as to characterize the relationship between gepotidacin exposure and E. coli resistance amplification. Results showed that the gepotidacin free-drug AUC/MIC ratio was closely associated with changes in bacterial burden, and different AUC/MIC values were able to suppress E. coli growth and resistance development.

Summary: The study demonstrated that the combination of meropenem, cefepime, or aztreonam with Nacubactam exhibited potent in vivo antimicrobial activity in a murine thigh-infection model against Klebsiella pneumoniae and Escherichia coli, including carbapenem-resistant Enterobacterales and carbapenemase-producing Enterobacterales isolates. These findings suggest that these combinations are potential candidates for the treatment of CRE and/or CPE infections.

Summary: This study investigates the feasibility of using real-time quantitative polymerase chain reaction (RT-qPCR) to quantify bacterial DNA loads (BDLs) for assessing treatment efficacy of vancomycin in late-onset sepsis (LOS). Serial blood samples were collected from premature and very low birth weight (VLBW) newborns, and BDLs were measured using RT-qPCR. Population pharmacokinetic-pharmacodynamic modeling was performed to describe the relationship between vancomycin concentration and BDL elimination rate, and it was found that treatment response to vancomycin can be assessed as early as 8 hours after treatment initiation.
