Summary: In this study, inhibitors of EgDdls were screened and found to bind efficiently to the ATP binding pocket, forming stable interactions primarily through electrostatic and van der Waals interactions. These inhibitors slowed down the motion of the central domain and omega loop of EgDdls, inhibiting its function and potentially alleviating vancomycin resistance in enterococci.

Summary: A Clostridioides difficile strain lacking the ddl gene synthesizes D-Ala-D-Ser instead of D-Ala-D-Ala to replace it as an essential component of peptidoglycan. The constitutive expression of the vanG operon, regulated by vanS or vanR genes, leads to low-level resistance to vancomycin. However, the presence of D-Ala-D-Ala is required for efficient vancomycin induction of the vanG operon, indicating that vancomycin alone is unable to activate VanS. The coupling of vanG and ddl expression is mediated by the anti-activator role of D-Ala-D-Ser on DdlR.

Summary: Vancomycin is a potent and broad-spectrum antibiotic that binds to the d-Ala-d-Ala moiety of the bacterial cell wall. This study synthesized d-Ala-d-Ala silica gels for a new physicochemical screening method. The results showed that vancomycin can be purified using d-Ala-d-Ala silica gel.

Summary: A novel class of artificial metalloenzymes was synthesized and characterized based on the supramolecular interaction between vancomycin (Van), an antibiotic glycopeptide, and D-Ala-D-Ala (DADA) dipeptides. The type of link between DADA and the metal-chelating moiety was found to be crucial for inducing asymmetry in the reaction outcome, as highlighted by computational studies and catalytic results. In the reduction of quinaldine B, a significant 70% (S) enantiomeric excess was achieved using the [IrCp*(m-I)Cl]Cl subset of Van complex in 0.1 M CH3COONa buffer at pH 5.
