Summary: This study found that osteopontin (OPN) is highly expressed in gastric cancer cells and that OPN knockdown can significantly reduce the proliferation, invasion, and migration capabilities of these cells. Furthermore, OPN promotes the expression of MMP-2 and VEGF through the PI3K/AKT/mTOR signaling pathway, contributing to the increased proliferation, invasion, and migration of gastric cancer cells.

Summary: In this study, a combination therapy using ATRA and inhibitors against members of the PAM signaling pathway was found to significantly reduce MYC protein levels in AML cells. This combination treatment targeted MYC through independent mechanisms, effectively blocking proliferation and enabling differentiation.

Summary: The clinical benefit of monotherapy with PI3K/AKT/mTOR inhibitors in patients with advanced or recurrent ovarian cancer is limited to a small subgroup, and selecting patients based on current biomarkers did not significantly improve the response rate. Due to the substantial toxicity associated with this treatment, it is recommended that PI3K/AKT/mTOR inhibitors should be used cautiously and preferably in the context of clinical trials. Improved biomarkers are needed to better select patients for this treatment.

Summary: mTOR is involved in regulating energy metabolism, neuronal growth, insulin signaling, and autophagy, playing both beneficial and pathogenic roles in neurodegenerative diseases. Balanced actions of mTOR complexes may have implications for Alzheimer's disease, Parkinson's disease, Huntington's disease, Frontotemporal dementia, and Amyotrophic Lateral Sclerosis. Beyond rapamycin, rapalogs with improved tolerability and delivery modes hold promise in treating age-related conditions.

Summary: This study measured the potential beneficial effects of adding metformin and/or active vitamin D to the main cytotoxic drug used for treating colon cancer. The results demonstrated that metformin had superior anticancer effects and ameliorated the effects of chemotherapy. Additionally, the triple treatment with the drugs showed the best inhibition of cancer progression.

Summary: The mammalian target of rapamycin (mTOR), a signalling system, is necessary for various cell proliferation activities. It recognizes PI3KAKT stress signals as a serine-threonine kinase. The abnormal regulation of mTOR pathway has been proven to be crucial in cancer growth and advancement. This review primarily discusses the normal functions of mTOR as well as its abnormal roles in cancer development.
