Ethnic specificity of lupus-associated loci identified in a genome-wide association study in Korean women

Objectives To identify novel genetic candidates for systemic lupus erythematosus (SLE) in the Korean population, and to validate the risk loci for SLE identified in previous genome-wide association studies (GWAS). Methods We performed a GWAS in 400 Korean female SLE patients and 445 controls. Selected single-nucleotide polymorphisms (SNP) were then replicated in an independent cohort of 385 SLE patients and 583 controls (replication cohort 1), and in a further 811 SLE patients and 1502 controls (replication cohort 2). Results In the GWAS phase, rs9275428 located near HLA-DQB1 showed the strongest association with SLE (OR 0.50, false discovery rate (FDR) p=3.07x10(-6)). Although no loci reached genome-wide significance outside major histocompatibility complex (MHC), C8orf13-BLK, STAT4, CSMD1, DIAPH3, GLDC and TNFSF4 showed FDR p < 0.05. Our results suggest that STAT4, BLK, IRF5, PTTG1-miR-146a, UBE2L3 and TNFAIP3 are shared susceptibility loci among Caucasians and Asians, while ETS1, IKZF1, SLC15A4 are likely to be Asian-specific loci. In a combined analysis of 1596 SLE patients and 2540 controls for selected 22 candidate SNP, STAT4 and BLK as positive controls showed a strong association with SLE (FDR p=9.85x10(-13) and 2.28x10(-8), respectively). Of these, 16 candidates (PEX5L, TRAJ50, MYO18B, SOS1, ARHGAP26, SMURF1, CADPS, HAND1, FAM78B, DIAPH3, TBL1XR1, CSMD1, ZBTB20, C3orf21, HIPK1 and AP001042.1) showed only nominal significance (7.05x10(-4)<= FDR p <= 4.38x10(-2)). Conclusions There are similarities and differences in genetic susceptibility for SLE between Caucasian and Asian ethnic groups. Although 16 putative novel loci for SLE have been suggested in the Korean population, further research on a larger sample is required to discriminate truth from error.

Flare of adult-onset Still's disease following mRNA COVID-19 vaccination: a case report and review of literature

Summary: This study reports a case of a 49-year-old female who experienced a flare of adult-onset Still's disease (AOSD) after receiving the first dose of BNT162b2 mRNA COVID-19 vaccination. The patient presented with fever and nausea 4 days after vaccination and was ultimately treated with steroid pulse therapy. The study also discusses possible mechanisms linking vaccination with a flare of AOSD.

Recent advances in understanding the genetic basis of systemic lupus erythematosus

Summary: Systemic lupus erythematosus (SLE) is a polygenic chronic autoimmune disease with a high heritability of up to 66%, with most risk variants residing in non-coding regions. Recent studies have identified dozens of new SLE loci and integrated GWAS signals with other biological resources in genetic associations. The research also explores genetic risk scores for SLE and their associations with clinical features, as well as variant-driven pathogenetic functions and future challenges in genetic studies on SLE.

Summary: By combining genome-wide association studies and transcriptome-wide association studies, we identified candidate genes associated with systemic lupus erythematosus and discovered a strong association between the CD83 gene and SLE. We also identified 276 gene candidates at the 110 SLE loci and demonstrated a regulatory effect of a putative causal variant on the ACAP1 gene.

Clinical Features and Drug Retention of TNF Inhibitors in Older Patients with Ankylosing Spondylitis: Results from the KOBIO Registry

Summary: This study aimed to analyze the clinical features, outcomes, and reasons for discontinuing TNF inhibitor therapy in older patients with ankylosing spondylitis (AS). Data from a nationwide registry were used to compare clinical variables and outcomes. The study found that older patients had a higher incidence of hypertension and diabetes, and lower drug retention rates. However, improvements in disease activity and manifestations were comparable between older and younger patients.

Rho-dependent transcription termination proceeds via three routes

Increased Immunoglobulin Gamma-3 Chain C in the Serum, Saliva, and Urine of Patients with Systemic Lupus Erythematosus

Summary: This study evaluates the clinical value of Immunoglobulin gamma-3 chain C (IGHG3) by measuring and comparing its levels in different body fluids of patients with systemic lupus erythematosus (SLE). The levels of IGHG3 in saliva, serum, and urine were measured and analyzed in patients with SLE and healthy controls. It was found that IGHG3 levels were increased in the saliva, serum, and urine of patients with SLE, and correlated with certain clinical characteristics.

Transcriptional pause extension benefits the stand-by rather than catch-up Rho-dependent termination

Summary: In this study on bacterial Rho factor-dependent terminators, it was found that the three Rho-dependent termination routes operate compatibly and their efficiency depends on the termination pauses. The most abundant route involves Rho catching up with a paused RNA polymerase, leading to simultaneous release of transcript RNA and template DNA. The fastest route results in RNA-only release and recycling of RNAP, while the slowest route facilitates simultaneous releases. The termination efficiency of stand-by Rho positively correlates with pause duration, contrary to previous speculation.

Summary: Communication between CD4 T cells and cognate B cells is crucial for the maturation of GC-TFH cells and the induction of CD4 T cell-dependent humoral immune response. Mef2d regulates the formation of B:T synapse and the production of IL-21, thus controlling the differentiation of CD4 T cells into GC-TFH cells and the antibody production by B cells. The expression of MEF2D gene is inversely correlated with autoimmune parameters in SLE patients.

Current Status of Korean Alcohol Drinking in Accordance with the Korean Alcohol Guidelines for Moderate Drinking Based on Facial Flushing

Summary: Due to individual differences in alcohol metabolism, it is inappropriate to use a uniform standard for evaluating drinking status. In Korea, a guideline based on alcohol metabolism capabilities, as indicated by facial flushing response, has been established for moderate drinking. This study aimed to identify the drinking habits of Koreans according to this guideline. The results confirmed that approximately one-third of the population experienced facial flushing when drinking alcohol, and different drinking habits were observed within the same age and gender groups based on the presence of facial flushing. The accurate evaluation of drinking habits is challenging without data on facial flushing from large-scale studies or various medical examinations. It is crucial to ensure the confirmation of facial flushing during medical treatment or examinations to accurately evaluate drinking habits and prevent and resolve drinking problems.

Rosuvastatin treatment alone cannot alleviate lupus in murine model: a pilot study

Summary: This study aimed to evaluate the effect of rosuvastatin on inflammatory responses in lupus-prone mice. The results showed that rosuvastatin can decrease serum cholesterol levels and reduce the expression of inflammatory cytokines in lymphoid organs and kidneys. However, it did not improve clinical manifestations, autoantibody titres, or renal histopathology.

Translocating RNA polymerase generates R-loops at DNA double-strand breaks without any additional factors

Summary: In this single-molecule study, researchers discovered that actively transcribing RNA polymerase can act as an efficient sensor for DNA double-strand breaks (DSBs), resulting in the formation of R-loops. These findings provide valuable insights into the initiation of DSB repair on transcription templates across different organisms.

Effects of COVID-19 and Influenza Vaccination on Rheumatic Diseases: Results From a Survey of Patient- Reported Outcomes After Vaccination

Summary: This study compared the occurrence of adverse events and disease flares after COVID-19 and influenza vaccination in patients with autoimmune rheumatic diseases (ARDs). The results showed that the rates of adverse events and disease flares were significantly higher after COVID-19 vaccination than after influenza vaccination. Previous allergic reactions to other vaccines were the only factor associated with the occurrence of adverse events. There was no difference in the post-vaccine response between mixed and matched vaccines.
