Summary: This study conducted expression quantitative trait locus (eQTL) analysis using RNA-Sequencing to explore the link between genetic variants and gene expression in rheumatoid arthritis (RA) patients. The analysis identified 898 eQTL genes in synovium and blood, with 232 genes in common. A specific eQTL at HLA-DPB2 was found to be associated with clinical severity and the lympho-myeloid pathotype.

Summary: This study aimed to analyze the linear correlation between IL-1ra and IL-6 in patients with rheumatoid arthritis (RA) and their relationship with disease's inflammatory activity. IL-6 and IL-1ra levels were measured in 407 RA patients. No correlation was found between serum levels of IL-6 and IL-1ra. However, both cytokines were positively associated with disease activity and acute phase reactants. After controlling for covariates, disease activity scores were more strongly associated with IL-1ra compared to IL-6.

Summary: In this study, a machine learning-based computational method was used to automatically assess joint inflammation in rheumatoid arthritis (RA) through thermal imaging of the hands. The method, called Thermographic Joint Inflammation Score (ThermoJIS), was trained using data from patients with different types of arthritis and osteoarthritis, and evaluated based on patients with RA. The results showed that ThermoJIS correlated moderately with ultrasound scores and was able to detect joint inflammation even in patients in clinical remission. This opens up an opportunity to develop new tools for routine detection of joint inflammation.

Summary: Glioblastoma (GBM) is an incurable disease and personalized treatments are expected to be the key to therapeutic success due to the heterogeneity, complexity, and plasticity of GBM tumors. This study developed a 3D biosphere model using patient-derived cells (PDCs) from GBM and cancer-activated fibroblasts (CAFs) to mimic GBM progression and response to treatment. The co-culture of PDCs with CAFs showed similar growth and morphology to patients, and the response to standard therapy (Temozolomide and irradiation) was also consistent with GBM patients. This method provides a simple and reproducible way to obtain 3D cultures and has the potential for precision and personalized medicine.

Summary: Re-education of the tumor microenvironment with immune checkpoint inhibitors (ICI) has been a significant breakthrough in cancer treatment, but low response rates and immune-related adverse events (irAEs) remain challenges. A bispecific Nanofitin was engineered using anti-epidermal growth factor receptor (EGFR) and anti-programmed cell death ligand 1 (PDL1) modules to increase selectivity and safety. The study demonstrated that the bispecific Nanofitin elicited EGFR-directed PDL1 blockade, highlighting its potential in enhancing the safety and selectivity of PDL1 checkpoint inhibition.

Summary: Osteoarthritis is an inflammatory joint disease affecting cartilage, subchondral bone, and joint tissues. In this study, human adipose stromal cells were successfully encapsulated in alginate microgels and shown to retain their metabolic activity and ability to respond to inflammatory stimuli. When injected into a rabbit model of post-traumatic OA, the encapsulated cells exhibited properties matching those of non-encapsulated cells and showed a tendency toward decreased OA severity. These findings demonstrate the feasibility, safety, and efficacy of injecting cells encapsulated in microgels for long-term treatment of OA.

Summary: Consuming high dietary glucose and having hyperglycemia can lead to chronic complications. This study investigated the effects of long-term exposure to high glucose on the intestinal barrier. The results showed that high glucose induced morphological changes and disrupted the structure of junction proteins. It also affected the functionalities of the intestinal barrier, and these alterations were more pronounced in co-culture models.

Summary: This study reaffirms the relevance of simpler 3D culture methods, such as the Liquid Overlay Technique (LOT), for producing highly reproducible spheroids in the context of drug cytotoxicity measurements. The morphology of spheroids generated in round-bottom multiwell plates was found to be more repeatable than those generated in flat-bottom plates. The IC50 of doxorubicin on MNNG/HOS cells grown in 3D was significantly higher than in 2D cultures, indicating the potential of 3D models for drug response measurements.

Summary: Rheumatoid arthritis (RA) and periodontitis are chronic inflammatory diseases that lead to increased bone resorption. They have immunopathogenic similarities and a common inflammatory environment. Periodontal dysbiosis, triggered by periodontal pathogens, can promote or sustain bone resorption in inflamed joints in RA. Understanding the pathogenic mechanisms of osteoclasts and their precursors in these diseases could lead to the identification of new therapeutic targets for inflammatory bone resorption.
