Effects of Aging on Rheumatoid Factor and Anticyclic Citrullinated Peptide Antibody Positivity in Patients With Rheumatoid Arthritis

Summary: This study investigated the factors affecting RF and anti-CCP positivity in patients with RA. The results showed that age, sex, smoking habit, and BMI were associated with the positivity rates. Multivariable analysis revealed that age had an independent association with seronegative RA in female patients with no smoking history and BMI < 25.

Summary: The study demonstrated that monoclonal antibody 12G1 can directly detect citrullinated proteins in rheumatoid arthritis (RA) target tissue and sera, indicating its potential in arthritis induction. 12G1 may be useful for diagnosing RA, including seronegative RA, and could help to understand the pathophysiological role of citrullination in RA.

The diagnostic and prognostic value of IgG and IgA anti-citrullinated protein antibodies in patients with early rheumatoid arthritis

Summary: This study aimed to investigate the diagnostic performance of anti-CCP2 and anti-CCP3 IgG and IgA assays in patients with early RA, with a focus on the prognostic value of IgA ACPA. The results showed that the anti-CCP2 IgG and IgA assays had high specificity but low sensitivity. The anti-CCP3.1 assay had slightly higher sensitivity but lower specificity compared to the anti-CCP2 IgG assay.

Summary: This study found no significant association between the bacterial load of Porphyromonas gingivalis (P. gingivalis), a primary periodontal pathogen, and the levels of anti-cyclic citrullinated peptide (anti-CCP) antibodies in patients with rheumatoid arthritis (RA) and periodontitis. Despite the prevalence of periodontitis among RA patients, the bacterial load does not have an impact on the production of anti-CCP antibodies.

Summary: Although anti-citrullinated protein antibodies (ACPAs) are a hallmark of rheumatoid arthritis (RA), their in vivo functions remain unclear. In this study, monoclonal ACPAs derived from RA patients were expressed in mice and their functions were analyzed. One of the antibodies, clone E4, showed a protective effect against antibody-induced arthritis in mice, suggesting therapeutic potential for ACPAs in RA.

This work could shed light on the development of tolerogenic therapeutics as novel immunotherapies for rheumatoid arthritis.

The results showed that the four anti-CCP assays provided similar diagnostic information for rheumatoid arthritis (RA), with Qiangsheng's method being potentially more useful for large scale clinical screening due to its shorter testing time.

Treatment trials are needed to determine if thyroxine supplementation can improve the cardiovascular outcomes in these patients.

Treat-to-target in systemic lupus erythematosus: advancing towards its implementation

Summary: The treat-to-target (T2T) concept, which involves choosing a specific target, evaluating the outcomes, and taking action accordingly, has been successful in improving outcomes for patients with diabetes, hypertension, and rheumatoid arthritis. However, the application of this approach in systemic lupus erythematosus (SLE) requires measurable and achievable outcomes, as well as more therapeutic options. Currently, the available therapeutic options for SLE are limited, and there is a need for more effective and safer treatments. Fortunately, there have been significant advancements in clinical trial activity for SLE, especially with the development of novel therapies. With these expected advancements, it is likely that a wider range of treatments will be available for SLE in the near future, allowing for the routine implementation of a treat-to-target approach.

Work participation in patients with systematic lupus erythematosus: a systematic review

Summary: This systematic review identified variables associated with work participation outcomes in patients with systematic lupus erythematosus (SLE). Factors such as age, ethnicity, educational level, disease activity score, disease duration, disease manifestations, physical functioning, cognitive functioning were found to be predictors for work outcomes. It was also found that certain demographic and disease-related factors were associated with unfavorable work outcomes.

Summary: This study investigated the potential of whole-body PET/CT scanning with a macrophage tracer to image arthritis in patients with early RA. The results showed increased uptake of the macrophage PET tracer in the joints of clinically active early RA patients, especially in the feet. Quantitative PET measures showed moderate and significant correlations with clinical outcomes.

Summary: This study investigated the disease severity of COVID-19 in patients with rheumatic immune-mediated inflammatory diseases (IMIDs) and the development of SARS-CoV-2 antibodies in these patients. The study found that patients with IMIDs, especially those treated with B-cell targeting therapy, were more likely to be hospitalized when infected with SARS-CoV-2. However, treatment with other drugs did not have negative effects on the development of long-lasting humoral immunity against SARS-CoV-2.

Summary: Specific immunosuppressants may impair humoral responses after SARS-CoV-2 vaccination. Most patients receiving immunosuppressant therapy show similar humoral responses to controls after standard vaccination, although antibody titres may be moderately reduced. However, a third vaccination can enhance immune responses in patients with poor humoral responses.

Summary: The first outbreak of the COVID-19 pandemic resulted in the increased use of e-health technology in Dutch general practices. This study investigated the experiences of general practitioners and identified the necessary conditions for successful implementation of e-health. The findings indicate that while most GPs intend to continue using e-health applications extensively, the actual usage depends on various factors such as user characteristics.
