Summary: Colitis-associated colorectal cancer (CAC) is a severe complication of ulcerative colitis (UC), characterized by multiple lesions, worse pathological type, and poor prognosis. Macrophages, as innate immune cells, play a crucial role in both inflammatory response and tumor immunity. Enhanced macrophage infiltration in UC leads to the production of inflammatory cytokines, promoting UC tumorigenesis. Macrophage polarization into M1 has an anti-tumor effect, while M2 polarization promotes tumor growth. Targeting macrophages has shown effectiveness in preventing and treating CAC.

Summary: This study investigated the pathogenesis of ulcerative colitis (UC) and found that neutrophils may promote the conversion of UC into colitis-associated colorectal cancer (CAC). By analyzing transcriptome data, 65 differentially expressed genes related to UC were identified, as well as immune-related biological pathways. The association between neutrophils and UC was confirmed using CIBERSORT and WGCNA analysis. These findings were validated in a mouse model, and 5 genes were identified as potential biomarkers. The results contribute to a better understanding of the pathogenesis of CAC and provide new insights for its prevention and treatment.

Further research is needed to define the optimal follow-up strategy.

Summary: Immune checkpoint inhibitors (ICIs) can cause various immune-related adverse events, including cytokine release syndrome (CRS) and hemophagocytic lymphohistiocytosis (HLH). This study provides a systematic review of CRS and related side effects of ICI treatment, and highlights the rarity of these complications. The review also suggests the potential benefits of using IL-6 inhibitors in combination with ICIs to enhance antitumoral effects and reduce hyperinflammation.

Summary: Statistical prediction models are popular in applied research, but transferring the model to a different population poses a challenge. This study systematically evaluates various calibration approaches for two-class probability estimation. Logistic calibration and beta calibration consistently show the best results, with calibration on logit transformed probability estimates generally outperforming non-transformed estimates. Regression-based calibration approaches are recommended for validation studies.

Summary: This study examined the differences in content and form of titles between original research articles published in major medical journals between 2011 and 2020. The analysis of 500 randomly chosen articles revealed that there were minimal mentions of results, quantitative information, declarative titles, dashes, or question marks in the titles across all journals. However, the use of subtitles and methods-related items increased over time, while the use of phrasal titles decreased. The study also found variations in the use of study names among the journals, with the Lancet having the highest usage. The findings highlight the importance for authors to carefully consider the titles of journal articles in their target journal before submission.
