Cognitive reserve hypothesis: Pittsburgh Compound B and fluorodeoxyglucose positron emission tomography in relation to education in mild Alzheimer's disease

Objective: The reduced risk for Alzheimer's disease (AD) in high-educated individuals has been proposed to reflect brain cognitive reserve, which would provide more efficient compensatory mechanisms against the underlying pathology, and thus delayed clinical expression. Our aim was to find possible differences in brain amyloid ligand C-11-labeled Pittsburgh Compound B ([C-11]PIB) uptake and glucose metabolism in high- and low-educated patients with mild AD. Methods: Twelve high-educated and 13 low-educated patients with the same degree of cognitive deterioration were studied with PET using [C-11]PIB and F-18-fluorodeoxyglucose as ligands. The between-group differences were analyzed with voxel-based statistical method, and quantitative data were obtained with automated region-of-interest analysis. Results: High-educated patients showed increased [C-11]PIB uptake in the lateral frontal cortex compared with low-educated patients. Moreover, high-educated patients had significantly lower glucose metabolic rate in the temporoparietal cortical regions compared with low-educated patients. Interpretation: Our results suggesting more advanced pathological and functional brain changes in high-educated patients with mild AD are in accordance with the brain cognitive reserve hypothesis and point out the importance of development of reliable markers of underlying AD pathology for early AD diagnostics.

Differences in brain changes between adults with childhood-onset epilepsy and controls: A prospective population-based study

Summary: The study revealed that neurologic signs were more common in COE participants not in remission, particularly cerebellar signs. MRI white matter abnormalities were associated with absence of vocational education, and MRI hippocampal atrophy in COE subjects was associated with arterial hypertension. In the long-term follow-up, there were tendencies to accelerated brain aging in COE participants, especially in focal epilepsy.

Summary: The study evaluated the tracer uptake and lesion detectability of F-18-radiohybrid (rh)PSMA-7.3 in patients with prostate cancer. The results showed that F-18-rhPSMA-7.3 PET/CT demonstrated good detection of primary and metastatic PCa lesions, especially lesions with GS >= 4 + 3/GG >= 3.

Summary: This study examined the impact of education, age, and gender on cognitive screening for early Alzheimer's disease and found that education was the strongest predictor of cognitive performance. Summary: Bexmarilimab is a new humanized monoclonal antibody targeting CLEVER-1 and is currently being tested for macrophage-guided cancer immunotherapy. This study evaluated the biodistribution and stability of radiolabeled bexmarilimab in rabbits, and found that it rapidly cleared from the blood circulation and showed higher uptake in fibrotic renal cortex. These findings support future human PET studies for bexmarilimab treatment and disease monitoring.

Change in brain amyloid load and cognition in patients with amnestic mild cognitive impairment: a 3-year follow-up study

Summary: In the early stage of aMCI, the presence of amyloid pathology at baseline strongly predicted the accumulation of amyloid and decline in cognitive abilities during the follow-up period. Some patients remained amyloid negative at the end of the study without significant changes in F-18-Flutemetamol uptake or cognition.

Evaluation of renal glucose uptake with [18F]FDG-PET: Methodological advancements and metabolic outcomes

APOE ε4 gene dose effect on imaging and blood biomarkers of neuroinflammation and beta-amyloid in cognitively unimpaired elderly

Summary: This study used positron emission tomography (PET) and blood biomarker measurements to examine differences in neuroinflammation and beta-amyloid (Aβ) and their association in cognitively unimpaired individuals. The results showed that only Aβ burden measured by PET differed among cognitively unimpaired elderly with different APOE ε4 gene dose, while the differences in neuroinflammation markers were not significant. However, APOE ε4 gene dose seemed to modulate the association between neuroinflammation and Aβ.

Head-to-head comparison of plasma p-tau181, p-tau231 and glial fibrillary acidic protein in clinically unimpaired elderly with three levels of APOE4-related risk for Alzheimer's disease

Summary: Plasma phosphorylated tau (p-tau) and glial fibrillary acidic protein (GFAP) are both early markers of Alzheimer's disease (AD), reflecting different amyloid-beta-related processes. In clinically unimpaired elderly individuals, differences in p-tau levels were observed based on APOE4 gene doses, solely explained by brain amyloid-beta load. All plasma biomarkers correlated positively with amyloid-beta PET, with different spatial patterns for p-tau markers and GFAP. Only higher plasma GFAP levels were associated with lower cognitive scores.

The results suggest endocannabinoid dysregulation already in the preobesity state, as CB1R availability in abdominal adipose tissue was lower in the high-risk group and correlated with insulin sensitivity and unfavorable lipid profile.

Protective factors against school burnout symptoms in Finnish adolescents

Summary: Burnout rates among adolescents have increased significantly. This study examined coping strategies, resilience, and school atmosphere as predictors for burnout symptoms in adolescents. Results showed that problem-solving and seeking social support coping strategies were associated with fewer symptoms. A positive school atmosphere, particularly instilling meaning in students, had a similar relationship with burnout. However, resilience was unexpectedly associated with more symptoms, and females reported higher rates of burnout symptoms than males.
