Summary: Chitinase ChiC8-1 with high chitin-binding capacity was purified and characterized in this study. A modified affinity chromatography method was developed to purify ChiC8-1 and hydrolyze chitin simultaneously. This process simplifies the purification steps and enables direct production of chitin oligosaccharides.

Summary: This study assessed the toxicity of lufenuron on Spodoptera frugiperda and evaluated its effects on the growth and development of the insects. The results showed that lufenuron has high insecticidal activity against S. frugiperda and can significantly prolong the larval developmental duration and reduce the rates of pupation and emergence. Gene expression analysis revealed that lufenuron treatment can significantly reduce the expression of SfCHT gene, while the expression of SfCHS gene remained relatively stable. Moreover, lufenuron strongly interacted with chitinase and chitin synthase enzymes.

Summary: In this study, we developed a reliable method for chitin detection using a luciferase-fragment complementation assay. The use of a CatD mutant allowed for higher sensitivity and structure-specific detection of chitin. Furthermore, a sandwich ELISA using modified CatD showed a low detection limit for soluble chitin.

Summary: Partially acetylated chito-oligosaccharides (paCOSs) are bioactive compounds with potential medical applications, and their biological activities are largely dependent on their structural properties, especially the degree of polymerization (DP) and the position of acetyl groups. A newly discovered chitinase from Paenibacillus pabuli (PpChi) has been shown to produce structurally defined paCOSs with specific compositions, demonstrating stability and activity over a wide temperature range, making it a promising biocatalyst for industrial processes. When used in combination with a recently discovered fungal oxidase, PpChi significantly increases the release of oligosaccharides, contributing to the green production of valuable paCOSs with a well-defined structure.

Summary: Genome mining of pigmented Pseudoalteromonas has shown potential for bioactive compound and enzyme production. Proteomic analysis of Pseudoalteromonas rubra strain S4059 revealed an efficient chitin degradation system when grown on chitin. Deletion of GH19 chitinase in S4059 did not affect chitin degrading ability or secondary metabolite profile, suggesting its industrial potential.

Summary: This study aimed to develop a two-step chemoenzymatic method for producing short chain chitooligosaccharides. By pretreating chitin with various chemicals and enzymes, and optimizing the conditions, the production of chitooligosaccharides was increased. Furthermore, the purification of the products was achieved using cation exchange chromatography, laying the foundation for sustainable upcycling of coastal residual resources.

Summary: In this study, a broad-specificity and high stability chitinase (AfChi28) was identified and characterized from a marine fungus. The recombinant enzyme AfChi28 was expressed in Escherichia coli and purified for further analysis. It was found that AfChi28 is an acido-halotolerant- and temperature-resistant bifunctional enzyme with both endo- and exo-cleavage functions. The enzymatic products of AfChi28 mainly consist of GlcNAc and various chitooligosaccharides. The study also elucidated the catalytic mechanism and substrate-binding mode of AfChi28 through molecular docking. This research provides insights into the application and catalytic mechanism of chitinases from marine fungi.

Summary: The present study investigated the effects of chitosan with different molecular weights on the biomolecule profile of cell walls and membranes in Eruca vesicaria ssp. sativa leaves using FT-IR spectroscopy. The results showed that the molecular weight and concentration of chitosan influenced membrane instability, lipid peroxidation, and lignin production. Among the treatments, the 10 kDa chitosan at 5 mg L-1 was found to be the most effective.

Summary: The study evaluated the association between G6PD mutations and hemolytic anemia incidence in the Saudi population, identifying Mediterranean mutation as the most common. Three females with G6PD variant II suffered from severe hemolytic anemia, highlighting the importance of genetic variation in G6PD genes in the etiology of hemolytic anemia.
