Summary: The skin is populated by various immune cells and sensory neurons, and the interaction between the nervous and immune systems plays a crucial role in inflammatory skin diseases. Neurotransmitters secreted by nerve terminals mediate immune cell function, while immune factors derived from immune cells interact with neurons to induce itching. This review explores the effector functions of neurons on skin immune cells in mouse models of atopic and contact dermatitis, and discusses the contribution of specific neuronal subsets and immune factors to itch induction and inflammation. The findings also shed light on treatment strategies and the relationship between scratching and dermatitis.

Summary: Parental history of allergic diseases, exposure to smoking, limited diet diversity, contact with antibiotics, and lifestyle factors may increase the risk of allergies, while household exposures, air pollution, and socioeconomic status may act as risk factors. Understanding the relationship between these factors can help to better understand and prevent the development of allergic diseases.

Summary: This study investigated the association between cataract and allergic diseases. The findings suggest that individuals with allergic diseases are more likely to develop cataract compared to those without allergies. This association is particularly significant in male patients under the age of 50.

Summary: The understanding of the pathologic mechanisms of asthma and atopic dermatitis has improved in recent years. However, the high heterogeneity of both diseases complicates diagnosis and treatment, with current strategies mainly focusing on symptom control rather than full healing. There is a need for more personalized approaches to improve diagnosis and treatment efficacy.

Summary: This study explored factors that may modify associations of early-life animal exposure with asthma and allergic disease, and found that type of animal, source of exposure, parental history of asthma or allergy, and timing of exposure can modify these associations. It is important to consider these factors when assessing the risks associated with early-life animal exposure.

Summary: This study aimed to validate three models for predicting disease outcomes in paediatric ulcerative colitis. The results showed that the PROTECT model had good predictive accuracy for steroid-free remission at 3 months, but not at 12 months. The other two models did not achieve sufficient accuracy, far from the results reported in the original studies. This highlights the necessity for external validation of any prediction model prior to its implementation in clinical practice.

Summary: The study found that CXCR4 and its ligand CXCL12 were significantly upregulated in the skin of humans with AD through global transcriptomic and proteomic analyses. Skin-resident CXCR4(+) NKT cells were enriched and consistently elevated in models of mice with AD, suggesting their potential role in contributing to AD pathogenesis.
