Summary: Studied the dynamics of single stranded DNA in complex coacervate droplets and micelles. Found that the diffusion coefficient of DNA depends more strongly on DNA length than predicted by traditional models, which can be partly explained by changes in complex coacervate density and other factors. Also discovered that chain length polydispersity is the main cause of a broad distribution of exchange rates in micelles, and dye interactions significantly affect micelle exchange rates.

Summary: Fluorescence lifetime imaging microscopy (FLIM) with phasor analysis allows for easy visualization and analysis of fluorophores' lifetimes, but typically suffers from low photon budgets leading to poor signal to noise ratios. Traditional median filters used in phasor analysis degrade high spatial frequency FLIM information, especially edges and puncta. A new filtering strategy using complex wavelet filtering and Anscombe transform preserves fine structures and accurate lifetimes in photon starved FLIM imaging, outperforming median filters and enabling FLIM imaging with lower laser power and faster imaging speed.

Summary: The dynamics and structural characteristics of the PMAA-R123 and its interpolymer complex with PVP were investigated using single molecular fluorescence studies. Different environments experienced by the fluorophore under acidic pH resulted in distinct fluorescence anisotropy decay behaviors, while hydrolysis of the fluorophore under basic conditions led to its release from the polymer backbone. These complexes have potential for drug-delivery systems under physiological conditions.

Summary: Single molecule localization microscopy offers high-resolution imaging in structural biology, but incomplete fluorescent labelling limits its value. This paper presents a particle fusion approach to complete missing information and accurately determine the positions of fluorescent labels using Artificial Intelligence. The results show a good match between fluorescent and cryo-EM data.

Summary: The dynamics of condensed matter can be studied using the time-dependent Stokes shift of a fluorescent probe. The time-dependent spectral correlation function is described by spectral relaxation correlation times that characterize the relaxation processes around the excited-state probe. Phasor plot is a powerful tool to analyze time and frequency-domain data, providing spatial maps of spectral dynamics in complex structures like living cells. The analysis workflow also addresses the extraction of initial and relaxed generalized polarization and improved methods for model discrimination and parameter extraction in complex spectral relaxation phenomena.

Summary: The presence of microplastics in Manila clams from the Korean market was studied. Microplastics of various shapes, mainly composed of polystyrene, were detected. There was a weak correlation between the concentration of microplastics and the physical attributes of the clams. The estimated intake of microplastics by the Korean population through consuming Manila clams was substantial.

Summary: The intermolecular interactions between the polycation pTMAEMC and anionic xanthene dyes OG and EB in buffered aqueous solution were characterized using optical spectroscopic methods. Visible absorption spectroscopy showed the formation of ground-state pTMAEMC-dye complexes, and Benesi-Hildebrand analysis quantified the strength of the binding interactions. The ionic strength of the solution's sodium acetate buffer was found to affect the binding affinity of pTMAEMC for the dyes, with EB exhibiting stronger binding. Fluorescence spectroscopy and fluorescence correlation spectroscopy provided complementary data on the interactions, demonstrating fluorescence enhancements and quenching dependent on buffer concentration. Excited-state lifetime analysis revealed different interaction modes between pTMAEMC and the dyes. The conclusion of the study was that the polyelectrolyte effect of pTMAEMC, influenced by the buffer concentration, affects its binding profile with the dyes.

Summary: The sensitive and accurate detection of biomarkers is crucial in clinical diagnosis and drug discovery. Existing amplification-based methods face challenges of limited reproducibility and high background noise. To solve these problems, a robust plasmonic nanoparticle-coupled single-molecule kinetic fingerprinting (PNP-SMKF) method is developed, allowing ultrasensitive detection of protein kinase A (PKA). This method shows great potential as a versatile platform for enzyme detection and inhibitor screening.

Summary: The real-time kinase assay method using a ratiometric fluorescence probe can be applied to various small-molecule kinases. It tracks the reversible interchange of ATP and ADP, facilitating monitoring of phosphorylation and dephosphorylation in small-molecule kinases. The fluorescence change can be observed with a simple light source by the naked eye, making it a convenient method for enzyme activity analysis.

Summary: This study aimed to investigate the loading of sildenafil citrate in three commercial transdermal vehicles using 3D full-thickness skin equivalent and compare the results with permeability experiments using porcine skin. The results showed that the results obtained using the 3D skin equivalent were comparable to those obtained using porcine skin, suggesting that the 3D skin model can be a valid alternative for ex-vivo skin absorption experiments.
