Summary: This study reveals that the high concentration of pyruvate in the culture medium can affect the stability of HIF-1α, and NADH serves as a signal for the degradation of HIF-1α under chronic hypoxia through the SIRT1 and VHL signaling pathway. This degradation leads to a reduction in hypoxia-enhanced invasion and angiogenic activities. Additionally, the study demonstrates that VHL is involved in the degradation of HIF-1α through recognition of hydroxylation in normoxia and acetylation in hypoxia.

Summary: This study investigated the effect of HSYA treatment on autophagy in a rat model of ischemic stroke (IS). The results showed that HSYA treatment significantly alleviated IS-induced neurobehavioral deficits and reduced brain infarct area and tissue damage. HSYA inhibited post-IS autophagy induction in the brain by suppressing HIF-1α, BNIP3, and Notch1 expression. HSYA may have utility as a post-IS neuroprotective agent.

Summary: Metabolic adaption drives microglial inflammatory responses, and lactate shapes immunological and inflammatory states. In this study, the researchers investigated the involvement of lactate in regulating microglial inflammatory responses after cerebral ischemia. They found that lactate treatment inhibited the increase in inflammatory markers and NF-kappa B pathway induced by oxygen-glucose deprivation in BV2 cells. Lactate also activated HIF-1 alpha, which played a role in the protective response of microglia. Furthermore, lactate-treated BV2 cells improved the viability and reduced the apoptosis of neurons after ischemia. These findings suggest that lactate can modulate microglial inflammatory responses and promote neuronal survival.

Summary: Breast tumor cells recruit bone marrow-derived mesenchymal stem cells (BM-MSCs) and alter their cellular characteristics to establish a tumor microenvironment. BM-MSCs enhance tumor angiogenesis through various mechanisms. Our study found that the increased expression of HIF-1 alpha in BM-MSCs under breast tumor conditions is mediated by ROS and JAK/Stat3. This increased expression promotes the angiogenic sprouting capacity of endothelial cells through a VEGF-dependent mechanism.

Summary: The transcription factor hypoxia-inducible factor-1 alpha (HIF-1 alpha) has two transcriptional activation domains (NTAD and CTAD), and the exact effects of HIF-1 alpha CTAD in kidney diseases are poorly understood. Using HIF-1 alpha CTAD knockout mice, two mouse models of hypoxia-induced kidney injury were established. It was found that HIF-1 alpha CTAD(-/-) aggravated kidney injury and could transcriptionally regulate HK2, providing a potential therapeutic strategy for hypoxia-induced kidney injury.
