Summary: CTCF is a critical regulator of genome architecture and gene expression. This study shows that R-loops and G-quadruplex structures facilitate CTCF binding and regulate chromatin organization. The stability of R-loops and G-quadruplex structures can affect CTCF occupancy and gene expression.

Summary: G-quadruplexes (G4s) are noncanonical nucleic acid structures that have gained increasing recognition in recent years, particularly in virology. G4 ligands are known to target viral transcription and replication, and both viral and cellular G4s may be potential targets for antiviral therapeutics. This review covers various aspects of G-quadruplexes in both host and viral contexts, highlighting the importance of studying G4s for future antiviral drug discovery.

Summary: This study reveals the enrichment of G4 motifs near splice junctions, suggesting their regulatory role in RNA stability and splicing. G4 motifs play important roles in exon skipping and inclusion, and are associated with multiple RNA binding proteins. Furthermore, G4 motifs show strong enrichment at splice sites in mammals and birds, indicating the evolutionary conservation of this splice regulatory mechanism.

Summary: This study investigates the adaptability of left-handed G4 structures towards the presence of bulges, presenting the structural characteristics of left-handed G4s accommodating varying numbers of bulges. Bulges in left-handed G4s exhibit distinct features compared to those in right-handed G4s, and elucidation of the intricate structural details may enhance understanding of the potential roles and limitations of these unique structures.

Summary: G4access is a nuclease-based method for identifying DNA G-quadruplex forming regions genome-wide in open chromatin using sequencing. It enriches for computationally predicted G-quadruplexes and allows measurement of variations in G-quadruplex repertoire usage following treatment or inhibition. The study highlights the association of G-quadruplexes with open chromatin, transcription, and DNA methylation, and provides a new tool for studying G4s in cellular dynamics.

Summary: Monkeypox virus (MPXV) is a DNA virus endemic in West and Central Africa. Recent outbreaks in non-endemic nations have raised concerns, and the availability of effective treatment options is limited. In this study, the authors explored the potential of G-quadruplexes as therapeutic targets. They identified conserved putative quadruplex-forming sequences in MPXV isolates and demonstrated their formation using spectroscopy and scattering techniques. The authors also found that specific G4-binding partners and a small molecule called TMPyP4 interacted with MPXV G-quadruplexes and reduced the expression of MPXV proteins. These findings provide valuable insights for the development of therapeutics.

Summary: G-quadruplexes, which are nucleic acids containing quartets of guanines, play a role in regulating gene transcription. Helquat-based compounds have been identified as a new class of anti-HIV-1 inhibitors that can stabilize G-quadruplexes in the HIV-1 long-terminal repeat sequence. This discovery can contribute to the rational design of inhibitors targeting G-quadruplexes in HIV-1.

Summary: Repeat expansions are the main genetic cause of various neurodegeneration diseases. G-rich repeat sequences, such as CGG and GGGGCC, form functional G-quadruplexes and play important roles in bioprocesses. This review provides an overview of the contribution of G-quadruplex in repeat expansion disorders and summarizes the related mechanisms, including genetic instabilities, toxic RNA foci, and protein/peptide dysfunction.

Summary: Parasitic helminths infecting humans and cattle have significant impacts on health and livestock production. This study identified and compared G4 sequences in the genomes of various helminths and found that a Nematode species, Ascaris lumbricoides, had a high enrichment of stable G4 structures. Experimental confirmation of G4 formation and the discovery of small molecules that can selectively recognize and bind G4 motifs in a parasitic helminth demonstrate the potential for targeting G4 structures in the treatment of helminth infections.
