Concentration-Dependent Self-Assembly of an Unusually Large Hexameric Hydrogen-Bonded Molecular Cage

Severin Merget, Lorenzo Catti, Shani Zev, Dan T. Major, Nils Trapp, Konrad Tiefenbacher

Summary: A large hexameric cage based on intermolecular amide-amide dimerization is reported, with a diameter of 2.3 nm and a cavity volume of 2800 Å(3), larger than any previously known capsule/cage structure relying solely on hydrogen bonds. The self-assembly process in chlorinated organic solvents is strongly concentration-dependent, with the monomeric form prevailing at low concentrations. Additionally, the formation of host-guest complexes with fullerenes (C-60 and C-70) was observed.

Thioderivatives of Resorcin[4]arene and Pyrogallol[4]arene: Are Thiols Tolerated in the Self-Assembly Process?

Suren J. Nemat, Konrad Tiefenbacher

Summary: Three novel thiol-bearing resorcin[4]arene and pyrogallol[4]arene derivatives were synthesized and studied for their properties in self-assembly, disulfide chemistry, and Bronsted acid catalysis. The findings showed that one compound could form a hexameric hydrogen bond-based capsule in self-assembly, another compound could be covalently linked through disulfides, and the increased acidity of aromatic thio-substituent was not sufficient to replace HCl as a cocatalyst for capsule-catalyzed terpene cyclizations.

Expanding the Protecting Group Scope for the Carbonyl Olefin Metathesis Approach to 2,5-Dihydropyrroles

Fabian Huck, Lorenzo Catti, Gian Lino Reber, Konrad Tiefenbacher

Summary: Chiral pyrrolidine derivatives are crucial for natural product synthesis, and carbonyl olefin metathesis is a powerful tool for constructing these building blocks. The supramolecular resorcinarene catalyst enables access to chiral 2,5-dihydropyrroles under Bronsted acid catalysis, even tolerating Lewis-basic protecting groups. The product yield and selectivity in this catalytic system depend on the size of the substrates due to reaction occurring inside a closed cavity.

Xanthene[n]arenes: Exceptionally Large, Bow Shaped Macrocyclic Building Blocks Suitable for Self-Assembly

Jonathan Pfeuffer-Rooschuez, Lucius Schmid, Alessandro Prescimone, Konrad Tiefenbacher

Summary: A new class of macrocycles called xanthene[n]arenes has been synthesized, featuring a conformationally restricted bowl shape that allows for the synthesis of cavitands and self-assembly into molecular capsules via noncovalent interactions. The derivatization potential of these novel macrocycles has been demonstrated, showcasing their ability to serve as a new set of macrocyclic building blocks.

Optimized iminium-catalysed 1,4-reductions inside the resorcinarene capsule: achieving >90% ee with proline as catalyst

Daria Sokolova, Konrad Tiefenbacher

Summary: This study optimized the reaction conditions for iminium-catalysed 1,4-reductions within a supramolecular resorcinarene capsule, achieving enantioselectivities of up to 92% ee. Additionally, it was discovered that alcohol additives play a dual role in slowing down background reactions and increasing the rate of capsule-catalysed reactions.

Understanding the binding properties of phosphorylated glycoluril-derived molecular tweezers and selective nanomolar binding of natural polyamines in aqueous solution†

Michael Heilmann, Melina Knezevic, GiovanniMaria Piccini, Konrad Tiefenbacher

Summary: A modular synthetic platform for constructing flexible glycoluril-derived molecular tweezers was developed and investigated for their binding properties towards 16 organic amines through H-1 NMR titration. The results demonstrated that certain structural modifications can lead to potent and selective binding of natural polyamines by the molecular tweezers.
