Summary: By analyzing the metabolic profile and metabolism-related genes in Frem2 mutant mice embryos, differentially expressed genes associated with amino acid and energy metabolism were identified. KEGG metabolic analysis suggested that ATP binding cassette transporters and amino acid biosynthesis pathways may be involved in the pathogenesis of cryptophthalmos.

Summary: Spindle transfer (ST) is a potential method to prevent the transmission of mitochondrial DNA mutations. This study used single-cell triple omics sequencing to evaluate the safety of ST and found that it is generally safe for embryonic development, with only a minor delay in the DNA demethylation process at the blastocyst stage.

Summary: This study aimed to investigate the biometric parameters of the uterus in fetuses with anencephaly and compare them with normocephalic fetuses. The findings revealed significant differences in the uterus length and width between the anencephalic group and the control group. However, there were no differences in the growth curves of uterine width and length during the studied period.

Summary: This study reveals that DNA replication stress in human preimplantation embryos predisposes gene-poor regions to fragility, resulting in the formation of aneuploidies.

Summary: This study demonstrates the regulatory role of RUNX3 in normal human erythropoiesis. RUNX3 has a modest impact on early erythroid growth and development, but promotes growth and inhibits terminal differentiation in late-stage erythroid development. Overexpression of RUNX3 increases colony formation in liquid culture.

Summary: Studies on cultured embryos have provided insights into human peri-implantation development, but the detailed knowledge of peri-implantation lineage development and mechanisms remains unclear. By using 3D-cultured human embryos, the researchers have created a complete cell atlas of early post-implantation lineages, deciphering the composition and gene signatures of epiblast and hypoblast derivatives. They have also developed an embryo-like assembloid that can simulate the development of epiblast and hypoblast. Their findings shed light on human peri-implantation development and provide a useful model for understanding human embryogenesis.

Summary: Selecting the best embryo for embryo transfer is crucial in IVF treatment, and can be facilitated by methods like preimplantation genetic testing (PGT) or cultivation in a time-lapse system. Various morphological and kinetic parameters can predict the implantation potential of embryos, aiding in the selection process for greater success in clinical pregnancy outcomes.

Summary: Based on high-resolution imaging techniques, the morphogenesis of the left atrium was studied in human embryos and fetuses. The left atrium wall has multiple origins and the position of the pericardial reflections determines the borders of the pericardial cavity. The four pulmonary veins enter the dorsal left atrium tangentially, with the right pulmonary veins entering at a greater angle than the left pulmonary veins.

Summary: This study found that during the process of embryo culture, the number of aneuploid cells significantly decreased and the proportion of healthy embryos significantly increased. Additionally, mitochondrial mutations may lead to decreased mitochondrial function and affect embryonic development. Therefore, analyzing the number of mitochondrial mutations may be a novel method to select better mosaic embryos for transfer.

Summary: The role of RUNX3 in normal myeloid development and leukemia, particularly acute myeloid leukemia (AML), is investigated. The study shows that increased expression of RUNX3 in AML can contribute to the developmental arrest characteristic of the disease. Overexpression of RUNX3 inhibits myeloid differentiation and proliferation in human hematopoietic stem and progenitor cells (HSPC), while RUNX3 knockdown does not impact myeloid growth and development. These findings suggest that dysregulation of RUNX3 may play an important role in the pathogenesis of AML.

Summary: This study aimed to establish normal fetal size ranges of optic chiasm, optic nerves, and optic tracts using ultrasound and compare them with fetuses with anophthalmia/microphthalmia. Normal widths increased linearly with gestational age while optic nerve width in affected fetuses was significantly smaller. This suggests fetal anophthalmia/microphthalmia may be associated with optic nerve hypoplasia.

Summary: The development of sympathetic trunks in human embryos was visualized using 3D-reconstruction software, revealing a change in position from lateral to dorsolateral as a criterion to distinguish primary and secondary trunks. The differential growth accounted for the regional differences in position of the sympathetic trunks as they transitioned from para-aortic to prevertebral or paravertebral positions during development.

Summary: This study reveals that DNA damage occurs during embryonic DNA replication in human embryos, leading to chromosome breaks and aneuploidy. DNA damage primarily occurs in G2 phase cells and affects mitotic progression and blastocyst quality. The study also demonstrates slowed replication and asymmetry in replication forks during DNA replication.
