Toward a Predictive Model of Alzheimer's Disease Progression Using Capillary Electrophoresis-Mass Spectrometry Metabolomics

Alzheimer's disease (AD) is the most prevalent form of dementia with an estimated worldwide prevalence of over 30 million people, and its incidence is expected to increase dramatically with an increasing elderly population. Up until now, cerebrospinal fluid (CSF) has been the preferred sample to investigate central nervous system (CNS) disorders since its composition is directly related to metabolite production in the brain. In this work, a nontargeted metabolomic approach based on capillary electrophoresis-mass spectrometry (CE-MS) is developed to examine metabolic differences in CSF samples from subjects with different cognitive status related to AD progression. To do this, CSF samples from 85 subjects were obtained from patients with (i) subjective cognitive impairment (SCI, i.e. control group), (ii) mild cognitive impairment (MCI) which remained stable after a follow-up period of 2 years, (iii) MCI which progressed to AD within a 2-year time after the initial MCI diagnostic and, (iv) diagnosed AD. A prediction model for AD progression using multivariate statistical analysis based on CE-MS metabolomics of CSF samples was obtained using 73 CSF samples. Using our model, we were able to correctly classify 97-100% of the samples in the diagnostic groups. The prediction power was confirmed in a blind small test set of 12 CSF samples, reaching a 83% of diagnostic accuracy. The obtained predictive values were higher than those reported with classical CSF AD biomarkers (A beta 42 and tau) but need to be confirmed in larger samples cohorts. Choline, dimethylarginine, arginine, valine, proline, serine, histidine, creatine, carnitine, and suberylglycine were identified as possible disease progression biomarkers. Our results suggest that CE-MS metabolomics of CSF samples can be a useful tool to predict AD progression.

Neuroprotective potential of terpenoid-rich extracts from orange juice by-products obtained by pressurized liquid extraction

Neuroprotective Potential of Thinned Peaches Extracts Obtained by Pressurized Liquid Extraction after Different Drying Processes

Summary: In this study, a pressurized liquid extraction method was optimized and applied to thinned peaches dried under different conditions to evaluate their neuroprotective potential. The extracts obtained from freeze-dried samples with 50% ethanol in water at 180 degrees C showed the best neuroprotective potential. The identified metabolites, including isoferulic acid, 4-methyldaphnetin, coniferyl aldehyde, and 3,4-dihydroxyacetophenone, were able to cross the blood-brain barrier and were positively correlated with the neuroprotective potential.

In Vitro Study of the Blood-Brain Barrier Transport of Natural Compounds Recovered from Agrifood By-Products and Microalgae

Summary: Agrifood by-products and microalgae are rich sources of bioactive compounds with neuroprotective properties, but their effectiveness is limited by the blood-brain barrier (BBB). In this study, green extracts from various sources were tested for their permeability across an in vitro BBB model. Results showed that neuroprotective compounds like terpenoids, phytosterols, and phenolic compounds had high permeability. These findings support the potential use of these natural extracts as functional ingredients in food supplements or nutraceuticals with neuroprotective claims.

Contribution of Phenolics and Free Amino Acids on the Antioxidant Profile of Commercial Lemon Verbena Infusions

Summary: This study investigated the antioxidant potential, phenolic profile, and free amino acid profile of lemon verbena infusions from different commercial brands. Various assays showed that the infusions exhibited high antioxidant activity, which was correlated with total phenolics and total flavonoid contents. Phenylalanine also appeared to have an impact on the antioxidant activity. Significant differences were found among the evaluated parameters for one of the commercial brands, possibly due to different geographical origins. Overall, lemon verbena infusions contain a variety of bioactive compounds with therapeutic potential.

Online Extraction Followed by LC-MS/MS Analysis of Lipids in Natural Samples: A Proof-of-Concept Profiling Lecithin in Seeds

Summary: Sample preparation is a complex and time-consuming procedure that directly affects analysis quality. Recent studies have explored online extraction coupled with liquid chromatography-mass spectrometry (OLE-LC-MS) as a promising alternative for analyzing analytes in complex solid matrices. In this study, the lecithin profile in various seeds was analyzed using OLE-LC-MS, demonstrating its feasibility without the need for offline solvent extraction. Optimization of the extraction phase gradient method and exploration of different analytical columns improved separation and repeatability. Overall, the OLE-LC-MS approach showed potential for lipidomics analysis in the future.

Additionally, new CE procedures for chiral separation, evaluating the effects of food processing, and recent developments in microchip CE and Foodomics applications are discussed.

Applicability and Limitations of a Capillary-LC Column-Switching System Using Hybrid Graphene-Based Stationary Phases

Bacterial Carotenoids: Extraction, Characterization, and Applications

Summary: Natural carotenoids have important applications in pharmaceutical, cosmetic, nutraceutical, and food industries. The use of bacteria for carotenoid production has gained much attention in recent years. This review summarizes the methods for extracting and identifying bacterial carotenoids and highlights the use of high-performance liquid chromatography-mass spectrometry (HPLC-MS) for analysis and identification. The potential commercial use of these carotenoids is also discussed.
