Nonlinear Signal Response in Electrospray Mass Spectrometry: Implications for Quantitation of Arsenobetaine Using Stable Isotope Labeling by Liquid Chromatography and Electrospray Orbitrap Mass Spectrometry

Isotope amount ratio measurements by electrospray ionization. mass spectrometry show large systematic biases. Moreover, the signal ratio response can vary nonlinearly with respect to the amount ratio depending on the concentration of the analyte or coeluting matrix components, among other things. Since isotope dilution relies inherently on the linearity of response, accurate quantitation is then more difficult to achieve. In this study, we outline a method to eliminate the quantitation errors due to the effects of the nonlinear signal response. The proposed approach is a hybrid of the method of standard additions and isotope dilution allowing correction for nonlinear trend. As a proof of concept, determination of arsenobetaine content in fish tissue was performed using liquid chromatography coupled with a linear quadrupole ion trap (LTQ) Orbitrap mass spectrometer. The nonlinear isotope dilution method could, in principle, be applied to correct isotope ratio measurement biases in popular relative quantitation methods of biomolecules such as stable isotope labeling by amino acids in cell culture (SILAC), isotope-coded affinity tag (ICAT), or isobaric tags for relative and absolute quantification (iTRAQ).

The study found significant variation in uranium isotope ratios among the three candidate UOC CRMs, with uncertainties of the proposed certified values superior to available reference materials.

From sea salt to seawater: a novel approach for the production of water CRMs

Interlaboratory comparisons of chemical measurements: Quo Vadis?

Towards single cell ICP-MS normalized quantitative experiments using certified selenized yeast

Characterization of vanillin carbon isotope delta reference materials

Summary: Stable carbon isotope ratio measurements are used to investigate the origin of vanillin. This study analyzed commercial vanillin samples and vanilla products to establish a reference range of carbon isotope delta values in different vanillin samples. Two synthetic vanillin reference materials, VANA-1 and VANB-1, were developed for calibration purposes. The certified carbon isotope delta values of VANA-1 and VANB-1 were determined through characterization and testing using nine reference materials. The results are traceable to the VPDB.

Calibration model averaging in chemical analysis: a case study for the method of standard additions

Summary: In this study, it is demonstrated that the method of standard additions can lead to unreliable results in chemical analysis when the linear calibration model is forced to fit the data. Even with a well-designed standard addition experiment, results can still be biased by 10% when relying solely on the linear model. The Joint Committee for Guides in Metrology (GUM-6) has recently stressed the importance of addressing uncertainty in measurement models, and this study shows how model averaging can offer a practical solution to account for model uncertainty in the method of standard additions.

Preparation of O-18-labelled azaspiracids for accurate quantitation using liquid chromatography-mass spectrometry

Summary: In this study, O-18-labelled AZA1, AZA2, and AZA3 were prepared by reacting with (H2O)-O-18 under acidic conditions. The reaction kinetics and sites of incorporation were analyzed using LC-HRMS/MS. The results showed that AZA1 and AZA3 had four exchangeable oxygen atoms. A stable isotopically labelled internal standard method was developed using O-18-labelled AZA1 and AZA3, which showed high accuracy and reproducibility in quantifying AZAs in shellfish matrices.

Se-82 Metabolically-Labeled Yeast as a Matrix-Matched Isotope Dilution Standard for Quantification of Selenomethionine

Summary: Selenized yeast is widely used as a source of selenium in dietary supplements and research due to its high bioavailability and formation of selenium-containing metabolites. Accurate quantitation of selenomethionine, the major form of selenium in selenized yeasts, is challenging and time-consuming. The National Research Council Canada has developed an 82-Se-enriched selenized yeast Certified Reference Material, SEEY-1, which can be used as a matrix-matched spike for the analysis of selenomethionine. SEEY-1 enables reliable analysis of Se amino acid content in under an hour compared to the conventional hydrolysis method.

Ode to Bayesian methods in metrology

Summary: Bayesian statistical methods are increasingly used in measurement science and offer unparalleled means to exploit all relevant and reliable measurement information. This review explores Bayesian concepts and methods in measurement science, providing practical examples of their application. The review also demonstrates that Bayesian methods are feasible using current modeling and computational techniques, and that measurement results obtained with Bayesian methods can be empirically validated.

Determination of the isotopic composition of copper in a certified reference material HICU-1 by MC-ICP-MS using gravimetric isotope mixture calibration

Summary: Isotopic analysis by mass spectrometry relies on certified reference materials (CRMs) to correct instrumental isotopic fractionation. The currently recognized reference material for copper isotope ratio, SRM976, is no longer available. In this study, an independent isotope ratio measurement of a high-purity copper CRM, HICU-1, was reported and its isotopic composition was shown to be nearly identical to that of NIST SRM976.

Determination of selenium-containing species, including nanoparticles, in selenium-enriched Lingzhi mushrooms

Summary: Mushrooms are considered a valuable food source due to their high protein and fiber and low fat content. This study showed that selenium-enriched Lingzhi mushrooms contained low levels of selenium, with selenomethionine being the most abundant form. The mycelium of these mushrooms showed significant selenium accumulation and the presence of selenium nanoparticles, which could have potential medicinal value.
