Summary: Cell surface receptors play a critical role in cell signaling and constitute a significant portion of human genes. A new method based on extracellular vesicles has been developed to detect receptor-ligand interactions in membranes, showcasing broad applicability and enhanced detection capability. This platform bypasses the need for membrane protein purification and allows for characterization of interactomes for any cell surface-expressed target in its native state.

Summary: In this study, we systematically mapped the direct protein interactions across a recombinant library that includes most of the surface proteins on human leukocytes, providing a high-confidence and quantitative view of the receptor wiring that connects human immune cells. By integrating our interactome with expression data, we identified trends in the dynamics of immune interactions and constructed a mathematical model that predicts cellular connectivity. We also developed an interactive multi-tissue single-cell atlas to infer immune interactions throughout the body and linked our receptor interactions to functional roles through protein stimulation and microscopy.

Summary: The study shows that RIP1 plays a crucial role in the onset and progression of osteoarthritis (OA) by mediating chondrocyte necroptosis and disrupting extracellular matrix (ECM) metabolism. Inhibition of RIP1 activity protects rats from cartilage degradation and limb pain, and bone morphogenetic protein 7 (BMP7) is identified as a downstream target that mediates RIP1-induced chondrocyte necroptosis and OA manifestations. This provides a new therapeutic target for OA.

Summary: This study aimed to investigate the effects of key pathogenic genes and extracellular matrix (ECM) on osteogenic differentiation injaw ameloblastoma (AB). Essential genes for AB were identified through whole-exome sequencing and microarray datasets analysis, and the expression of key genes and their encoded proteins in AB tissues was explored. ABdECM tissues were generated and the osteogenic development of PDLSCs was mimicked to simulate the AB tumor microenvironment. The study found that abnormal ECM proteins encoded by COL4A2, COL1A2, FBN1, and HPSE genes can cause disturbance in the ECM environment of AB and promote bone resorption.

Summary: This study comprehensively investigated the interactions between receptor activity-modulating proteins (RAMPs) and G protein-coupled receptors (GPCRs) using a bioluminescence resonance energy transfer (BRET) approach. The results revealed that RAMPs modulate the signal transduction of glucagon receptor family members in a receptor-specific manner. These findings provide new insights into the role of RAMPs in ligand recognition and receptor activation.

Summary: This study compared the results of different methods for measuring the amount of acetyl-Coenzyme A. The colorimetric ELISA kit did not produce interpretable results, while the fluorometric enzymatic kit showed comparable results to the LC-MS-based methods depending on the matrix and extraction conditions. LC-MS/MS and LC-HRMS methods produced well-aligned results, especially when using stable isotope-labeled internal standards.

Summary: This study investigates the co-catalytic capabilities of MoO3 nanosheets in enhancing the enzyme-like catalytic activity of a two-dimensional ultrathin Fe(III)-modified covalent triazine framework (Fe-CTF) under neutral pH conditions. The Fe-CTF/MoO3 co-catalytic system exhibits enzyme-mimicking activity and enables the development of a colorimetric method for glucose detection. Furthermore, a straightforward one-pot colorimetric method is established for screening XOD inhibitors.
