Summary: This study examined the comparability of cutoffs for Elecsys immunoassays for different measurements in cerebrospinal fluid (CSF) in predicting Alzheimer's disease (AD)-related outcomes. The results showed that CSF A beta 42/p-tau181 was more robust to measurement variability and may be the preferred measure in clinical practice and trials. The findings also indicated that the use of plasma A beta 42/40 led to a greater number of individuals falling within the gray zone.

Summary: This study aims to identify biomarkers for Alzheimer's disease (AD) and finds that the A beta 37/42 ratio can be used to distinguish AD from normal aging. Experimental results show that this ratio outperforms the traditional ratio in differentiating physiological and pathological states in cell culture, brain tissue, and cerebrospinal fluid. The findings may provide a new indicator for early diagnosis of AD.

Summary: Alzheimer's disease is a prevalent neurodegenerative disorder characterized by progressive deterioration, loss of neurons, and cognitive decline. Amyloid beta and Tau proteins are key biomarkers of the disease, and their abnormal levels in biofluids, particularly cerebrospinal fluid, are directly linked to the incidence and progression of Alzheimer's. Developing sensitive and reliable methods for screening and evaluating these biomarkers in cerebrospinal fluid can aid in early diagnosis and treatment. This article discusses the impact of Alzheimer's on cerebrospinal fluid and provides an overview of the latest biosensors for detecting amyloid beta and Tau in cerebrospinal fluid samples.

Summary: This study found that levels of A beta 42 and pTau in cerebrospinal fluid are more direct indicators of the pathogenesis of Alzheimer's disease than clinical diagnosis. Summary: This study investigated the association between cerebrospinal fluid biomarkers (p-tau and β-amyloid) and spatial cognition in clinically normal older adults. The results showed significant correlations between these biomarkers and spatial cognition scores on two tasks. Lower entorhinal cortical volume was associated with poorer performance on both tasks and accounted for 18%-22% of the direct association between p-tau and spatial cognition scores. The degeneration of the entorhinal cortex plays a significant mediating role in the association between p-tau and spatial assessments in cognitively normal adults.

Summary: This study investigated the relationship between A beta 42/A beta 40 ratios and cerebral A beta pathology using App knock-in mouse models. The results showed a negative association between A beta 42/A beta 40 ratios and cerebral A beta plaque burden in App(NL-F/NL-F) mice, with stronger correlations observed in CSF compared to serum. Additionally, the decline in A beta 42/A beta 40 ratios in CSF and serum occurred after the deposition of A beta aggregates in the brain reached significance. However, no similar changes were observed in App(NL/NL) mice.

Summary: According to the beta-amyloid (A beta) hypothesis, brain A beta accumulation is the primary cause of cognitive deficit and dementia in Alzheimer's disease (AD). While many anti-A beta drugs have failed in clinical trials, recent studies have shown encouraging results for antibodies that clear amyloid plaques. These findings suggest that decreased levels of soluble monomeric A beta may be the main driver of AD, rather than the aggregated forms.

Summary: Despite the long history of Alzheimer's Disease, many aspects of the disease, including its pathogenesis, clinical spectrum definition, and therapeutic approach, remain obscure. The commonly used biomarkers, A beta and phosphorylated Tau protein aggregates, have their limitations and biases. The definition of Mild Cognitive Impairment, which is often used for AD prediction, has changed over time, leading to biases in related studies. Plasma biomarkers and novel candidate molecules, such as microglia biomarkers, show promise for diagnosing and monitoring AD. New AD markers are urgently needed for identifying molecular targets for treatment strategies.

Summary: This study compared the results of different methods for measuring the amount of acetyl-Coenzyme A. The colorimetric ELISA kit did not produce interpretable results, while the fluorometric enzymatic kit showed comparable results to the LC-MS-based methods depending on the matrix and extraction conditions. LC-MS/MS and LC-HRMS methods produced well-aligned results, especially when using stable isotope-labeled internal standards.

Summary: This study investigates the co-catalytic capabilities of MoO3 nanosheets in enhancing the enzyme-like catalytic activity of a two-dimensional ultrathin Fe(III)-modified covalent triazine framework (Fe-CTF) under neutral pH conditions. The Fe-CTF/MoO3 co-catalytic system exhibits enzyme-mimicking activity and enables the development of a colorimetric method for glucose detection. Furthermore, a straightforward one-pot colorimetric method is established for screening XOD inhibitors.
