Optimization and validation of an alternative method to evaluate total reactive antioxidant potential

The total reactive antioxidant potential (TRAP) is one of the methods most employed to estimate the antioxidant capacity of samples in vitro. This method is based on the quenching of luminol-enhanced chemiluminescence derived from the thermolysis of 2,2'-azo-bis(2-amidinopropatie)dihydrochloride (AAPH) as the free radical source. However, this method can present limitations when the sample does not present a lag phase. In addition, there are no studies regarding TRAP assay validation. In this context, the aim of this work was to optimize and validate this method and to propose another evaluation method using the area under the curve (AUC). The main condition established was the need for the stabilization of the system, at 7000 s, before the addition of the antioxidant to be tested. Both evaluation methods were validated using Trolox (6-hydroxy-2,5,7,8-tetramethylchroman-2-carboxylic acid) as a calibrator in the range of 50 to 250 nM, and all parameters showed satisfactory results: specificity, linearity (r>0.99), precision (intra and interassay relative standard deviations <5%), robustness, and the limits of detection and quantitation (low and similar for both methods). The main advantage of the use of AUC is to evaluate the antioxidant potential of samples that do not present lag phase. (C) 2008 Elsevier Inc. All rights reserved.

Summary: In this study, the label-free mass spectrometry proteome of Erythroxylum pungens (roots, barks, leaves) harvested from its natural habitat was described. It was found that processes related to defense were most important in roots, and there were different main protective proteins among organs, indicating part-specific strategies of environmental adaptation.

Taxonomic significance of coumarins in species from the subfamily Mutisioideae, Asteraceae

Summary: Asteraceae, the largest family of angiosperms, has a subfamily called Mutisioideae which is known for producing 5-methylcoumarins, compounds that are rare in plants in general. These compounds are almost exclusively found in species of this subfamily within Asteraceae. The different tribes within Mutisioideae have variations in the substitution patterns of these compounds, suggesting taxonomic significance. Most of the 5-methylcoumarins in Mutisieae species have terpene moieties attached to the coumarin scaffold, while in species of Onoserideae and Nassauvieae tribes, sesquiterpene units are predominantly attached to the alpha-pyrone moiety. Furanocoumarins, another type of coumarin, are only found in Mutisieae species, which is rare in Asteraceae. The biological properties of 5-methylcoumarins have been poorly studied.

Summary: This study focused on Erythrina velutina, a Brazilian native tree, and identified it as a rich source of bioactive phenolics, particularly flavonoids. Through metabolic and proteomic analysis, as well as integration with transcriptome data, key genes and transcription factors involved in flavonoid biosynthesis were identified. The findings provide a foundation for understanding the molecular and metabolic responses in this medicinally important species.

Root exudation of oxalic acid in Lotus corniculatus in response to aluminum toxicity

Summary: The objective of this research was to investigate the existence of root exudation of organic acid in Lotus corniculatus germplasms subjected to toxic aluminum levels and its effect on the Al content in root tissue and plant development. The experiment showed that root exudation was closely related to reduced accumulation of Al in the root tissue. The genotype UF-T2, which is tolerant to Al, excreted 80% more oxalic acid in the presence of Al compared to other germplasms.

Summary: Tropane alkaloids are specialized plant metabolites found mainly in the Erythroxylaceae and Solanaceae families. Despite their structural similarities due to the tropane ring, they exhibit distinct pharmacological actions. Studies on the Erythroxylum species in Brazil, especially Erythroxylum pungens, have identified potential chemical markers like 3-(2-methylbutyryloxy)tropan-6,7-diol, which showed behavioral effects in zebrafish models. The alkaloid also demonstrated potential in affecting antidepressant pathways, highlighting its possible use in treating depression-like symptoms.

Pyrophosphate detection method using 5-Br-PAPS to detect nucleic acid amplification - Application to LAMP method

Summary: This study proposes a method for detecting nucleic acid amplification using pyrophosphate, which requires only two reagents and an automated analyzer. Comparison of colorimetric, fluorometric, and liquid chromatography-mass spectrometry assays for acetyl-coenzyme A

Summary: This study compared the results of different methods for measuring the amount of acetyl-Coenzyme A. LC-MS/MS and LC-HRMS methods produced well-aligned results, especially when using stable isotope-labeled internal standards.

A novel fusion technology utilizing complex network and sequence information for FAD-binding site identification
