Application of dried spot cards as a rapid sample treatment method for determining hydroxytyrosol metabolites in human urine samples. Comparison with microelution solid-phase extraction

Two different rapid sample pretreatment strategies, dried spot cards, and microelution solid-phase extraction plates (mu SPE), with ultra-high performance liquid chromatography coupled to tandem mass spectrometry (UPLC-MS/MS) have been developed and validated for the determination of hydroxytyrosol and its metabolites in spiked human urine samples. Hydroxytyrosol, hydroxytyrosol-3'-O-glucuronide, hydroxytyrosol-4'-O-glucuronide, hydroxytyrosol-3-O-sulphate, and homovanillic alcohol-4'-O-glucuronide were used as the target compounds. Using the FTA DMPK-A dried urine spot card under optimum conditions, with 5 mu L of preconcentrated urine volume and 100 mu L of methanol/water (50/50, v/v) as the elution solvent, the extraction recovery (%R) of the compounds studied was higher than 80 %, and the matrix effect (%ME) was less than 8 %. The stability of these cards and punching at the centre or side of the card were also studied, obtaining an excellent stability after 7 days of storage and complete homogeneity across the surface of the dried drop. The different mu SPE parameters that affect the efficiency were also studied, and under optimum conditions, the %R and the %ME were higher than 70 % and lower than 17 %, respectively. The linearity range in dried urine spot cards was 2.5-20 mu M for all the metabolites, with the exception of hydroxytyrosol-3-O-sulphate and hydroxytyrosol, which were 0.3-70 mu M and 2.5-50 mu M respectively. With regards to mu SPE, the linearity range was 0.5-5 mu M for all the studied compounds, except for hydroxytyrosol-3-O-sulphate, which was 0.08-5 mu M. The quantification limits (LOQs) were 0.3-2.5 mu M and 0.08-0.5 mu M in dried spot cards and in mu SPE, respectively. The two developed methods were then applied and compared for determining hydroxytyrosol and its metabolites in human 24 h-urine samples after a sustained consumption (21 days) of a phenol-enriched virgin olive oil. The metabolites identified were hydroxytyrosol in its glucuronide and sulphate forms, homovanillic alcohol in its glucuronide and sulphate forms, homovanillic acid sulphate and hydroxytyrosol acetate sulphate.

Summary: Hesperidin, a flavanone found in citrus fruits, has been reported to have health benefits. However, its bioavailability is low among individuals. This study aimed to evaluate the impact of micronization process and the ratio of 2R- and 2S-hesperidin diastereoisomers on hesperidin bioavailability. The results showed that micronized 2S-hesperidin extract had higher bioavailability compared to the standard mixture of 2S-/2R-hesperidin extract, likely due to the increased formation of hesperidin catabolites. Micronization process also improved hesperidin bioavailability.

Impact of Health and Social Factors on the Cardiometabolic Risk in People with Food Insecurity: A Systematic Review

Summary: Food insecurity is closely related to people's health and quality of life. This review explores the impact of food insecurity on cardiometabolic risk factors in adults in high-income countries. The findings suggest that food insecurity is significantly associated with cardiovascular risk factors. Integrating food insecurity screening and measurement of cardiometabolic risk factors into routine clinical care may help identify vulnerable populations and improve non-communicable disease prevention and management.

A comparative evaluation of the phenol and lycopene content of tomato by-products subjected to different drying methods

Summary: This study focused on dehydrating industrial tomato pomace using innovative techniques (microwave and spiral flash dryer) and conventional techniques (freeze-drying and hot air-drying) to produce ingredients rich in bioactive compounds. The effect of different drying methods on the bioactive composition of the tomato by-products was analyzed. Microwave dehydration improved the retention of flavanone-like compounds and lycopene, while spiral flash drying resulted in higher concentrations of flavonols and phenolic acids. These findings suggest that drying processes using microwave and spiral flash could be promising for producing high added-value ingredients from tomato by-products.

Bioavailability of Organosulfur Compounds after the Ingestion of Black Garlic by Healthy Humans

Summary: By using UHPLC-HRMS, this study identified and quantified 33 OSCs excreted in urine 24 h after the consumption of 20 g of black garlic. The main OSCs were methiin, isoalliin, S-(2-carboxypropyl)-L-cysteine, and S-propyl-L-cysteine. Metabolites such as NASAC, NASACS, and NACPC derived from SAC, alliin, and S-(2-carboxypropyl)-L-cysteine were also detected. The total excretion of OSCs after the ingestion of black garlic was 64,312 +/- 26,584 nmol.

The role of PPAR-gamma in memory deficits induced by prenatal and lactation alcohol exposure in mice

Summary: Patients with Fetal Alcohol Spectrum Disorder (FASD) exhibit long-lasting cognitive disabilities, including memory deficits, but the underlying neurobiological mechanisms are not clear. By studying alcohol-exposed mice, researchers found that prenatal and lactation alcohol exposure induces FASD-like memory impairments through the reduction of N-acylethanolamines (NAEs) and peroxisome proliferator-activated receptor gamma (PPAR-gamma) in the hippocampus during a childhood-like period. Pharmacological interventions targeting NAEs and PPAR-gamma were able to improve memory deficits, and overexpression of PPAR-gamma in hippocampal astrocytes mitigated memory impairments induced by alcohol exposure.

Olive Oil Phenolic Compounds' Activity against Age-Associated Cognitive Decline: Clinical and Experimental Evidence

Summary: Epidemiological studies suggest that consuming olive oil rich in phenolic bioactive compounds may lower the risk of neurodegenerative diseases and improve cognitive performance in the elderly. Preclinical evidence indicates that olive oil phenolic compounds can counteract oxidative stress and neuroinflammation, key processes in neurodegeneration. Clinical evidence suggests that high consumption of olive oil phenolic compounds is associated with better cognitive preservation, independent of dietary patterns. Further research is needed to understand the mechanisms and potential clinical applications.

Urinary excretion of organosulfur compounds after acute ingestion of black onion

Summary: This study monitored the excretion of organosulfur compounds (OSCs) in urine after acute ingestion of black onion. A total of 31 OSCs were detected, with the main components being methiin, isoalliin, and deoxypropiin. N-acetylated metabolites of the major OSCs were also found in urine. This study provides the basis for further research on the metabolism of OSCs after black onion consumption.

Assessment of human inter-individual variability of phloretin metabolites in urine after apple consumption. AppleCOR study

Summary: This study aimed to assess the inter-individual variation in phloretin absorption and metabolism and to seek possible phloretin metabotypes following apple snack consumption. The results showed that there is extensive inter-individual variability in the excretion of phloretin phase-II conjugates, which could be related to factors such as oral microbiota activity and lactase non-persistence trait. The study also found inconsistent effects on post-prandial serum glucose concentrations, with a tendency for decreases to be associated with higher excretion of phloretin phase-II conjugates.
