Chronic hepatitis C virus (HCV) infection is a global health issue. Although its progression is reported to be closely associated with lipids, the way in which the plasma lipidome changes during the development of chronic HCV infection in humans is currently unknown. Using an improved quantitative high-throughput lipidomic platform, we profiled 284 lipids in human plasma samples obtained from healthy controls (n = 11) and patients with chronic HCV infection (n = 113). The intrahepatic inflammation grade (IG) of liver tissue was determined by biopsy. Two types of mass spectrometers were integrated into a single lipidomic platform with a wide dynamic range. Compared with previous methods, the performance of this method was significantly improved in terms of both the number of target sphingolipids identified and the specificity of the high-resolution mass spectrometer. As a result, 44 sphingolipids, one diacylglycerol, 43 triglycerides, 24 glycerophosphocholines, and 5 glycerophospho-ethanolamines were successfully identified and quantified. The lipid profiles of individuals with chronic HCV infection were significantly different from those of healthy individuals. Several lipids showed significant differences between mild and severe intrahepatic inflammation grades, indicating that they could be utilized as novel noninvasive indicators of intrahepatic IG. Using multivariate analysis, healthy controls could be discriminated from HCV patients based on their plasma lipidome; however, patients with different IGs were not well discriminated. Based on these results, we speculate that variations in lipid composition arise as a result of HCV infection, and are caused by HCV-related digestive system disorders rather than progression of the disease.

Summary: The study demonstrated that ethanol extract of Aster tataricus (ATE) could significantly promote small intestinal transit, reduce fecal amount, increase fecal water content, and alleviate colonic pathological damage. In vitro experiments showed that ATE could attenuate constipation by antagonizing acetylcholine binding to muscarinic receptors, inhibiting calcium influx, and anti-inflammatory effects in a concentration-dependent manner.

Effects of motor skill level and speed on movement variability during running

Wei Wang, Feng Qu, Shangxiao Li, Liduan Wang

Summary: This study investigated the effects of motor skill level and running speed on movement variability, particularly focusing on stride length, joint angles, and coordination. Results showed significant interactions between skill level and speed on movement variability, with athletes demonstrating greater angle and coordination variability during sprinting. The relationship between coordination variability and performance was found to be complex, with more longitudinal studies needed to fully understand their connection.

Summary: The study found that Shengyu Decoction (SYD) can improve the cognitive abilities of rats with vascular cognitive impairment (VCI). Its active ingredients improve cognitive impairment by affecting the AKT/HIF-1 alpha/VEGF and p38 MAPK/NF-kappa B signaling pathways, promoting cerebrovascular generation, and ameliorating neuroinflammation.

Summary: The hepatotoxic equivalent markers (HEMs) of Polygonum multiflorum Thunb. (PMT) were identified as 2,3,5,4-tetrahydroxystilbene-2-O-beta-D-glucoside (TSG) and emodin-8-O-glucoside (EG). It was found that PMT induced liver injury by interfering with purine metabolism.

Summary: A strategy combining network pharmacological technique with integrated pharmacokinetics was developed to address the potential liver injury caused by Polygonum multiflorum. A HPLC-QQQ-MS method was established for the simultaneous quantification of five effective compounds. Pathway enrichment analysis revealed that potential liver injury is primarily associated with PI3K-Akt, MAPK, Rap1, and Ras signaling pathways.

Salidroside orchestrates metabolic reprogramming by regulating the Hif-1 alpha signalling pathway in acute mountain sickness

Xiaoning Yan, Jie Liu, Meixia Zhu, Lirong Liu, Yijun Chen, Yinhuan Zhang, Menghan Feng, Zhixin Jia, Hongbin Xiao

Summary: The study revealed that components of Rhodiola crenulata can induce metabolic reprogramming by regulating the Hif-1 alpha signaling pathway, activating compensatory responses. Microglia sensed low oxygen levels earlier than neurons, accompanied by elevated expression of Hif-1 alpha protein.
