Implementation of comprehensive two-dimensional gas chromatography-time-of-flight mass spectrometry for the simultaneous determination of halogenated contaminants and polycyclic aromatic hydrocarbons in fish

In the presented study, comprehensive two-dimensional gas chromatography coupled to time-of-flight mass spectrometry (GC x GC-TOFMS) was shown to be a powerful tool for the simultaneous determination of various groups of contaminants including 18 polychlorinated biphenyls (PCBs), seven polybrominated diphenyl ethers (PBDEs), and 16 polycyclic aromatic hydrocarbons (PAHs). Since different groups of analytes (traditionally analyzed separately) were included into one instrumental method, significant time savings were achieved. Following the development of an integrated sample preparation procedure for an effective and rapid isolation of several groups of contaminants from fish tissue, the GC x GC-TOFMS instrumental method was optimized to obtain the best chromatographic resolution and low quantification limits (LOQs) of all target analytes in a complex mixture. Using large-volume programmable temperature vaporization, the following LOQs were achieved-PCBs, 0.01-0.25 mu g/kg; PBDEs, 0.025-5 mu g/kg; PAHs 0.025-0.5 mu g/kg. Furthermore, several capillary column combinations (BPX5, BPX50, and Rxi-17Sil-ms in the first dimension and BPX5, BPX50, Rt-LC35, and HT8 in the second dimension) were tested during the experiments, and the optimal separation of all target analytes even of critical groups of PAHs (group (a): benz[a]anthracene, cyclopenta[cd]pyrene and chrysene; group (b): benzo[b]fluoranthene, benzo[j]fluoranthene and benzo[k]fluoranthene; group (c): dibenz[ah]anthracene, indeno[1,2,3-cd]pyrene and benzo[ghi]perylene) was observed on BPX5 x BPX50 column setup. Moreover, since the determination of target analytes was performed using TOFMS detector, further identification of other non-target compounds in real life samples was also feasible.

Detection of microscopic filamentous fungal biofilms - Choosing the suitable methodology

Summary: Microscopic filamentous fungi are adaptable microorganisms that can form biofilms on various surfaces. This study aimed to find a reliable method for measuring the biofilm growth of selected fungi strains. Multiple methods were tested, and crystal violet staining was found to be suitable for determining total biofilm biomass, while the MTT assay was suitable for determining its metabolic activity. Fluorescent stains successfully dyed the hyphae of microscopic fungi.

Assessment of organohalogenated pollutants in breast milk from the Czech Republic

Summary: This study conducted a biomonitoring survey on 231 human breast milk samples collected in 2019 and 2021 from two regions of the Czech Republic. The survey revealed the presence of 94 organohalogenated pollutants, including PCBs, OCPs, HFRs, PFAS, and PCNs. The concentrations of these pollutants were evaluated using advanced analytical techniques. The results showed a decline in the levels of banned POPs in Czech mothers, with higher concentrations of PCBs and OCPs observed in older breastfeeding primiparous women.

Toward building mass spectrometry-based metabolomics and lipidomics atlases for biological and clinical research

Summary: Current metabolomics and lipidomics studies have limitations in terms of the number of examined matrices, the breadth and scope of methods, reporting the number of metabolites, and data sharing. This article discusses the concept of metabolomics and lipidomics atlases, which characterize the quantitative distribution and relationships of metabolites in biological matrices and serve as a resource for future studies. The recommended approach includes combined sample extraction, a multiplatform mass spectrometry-based method, and the use of multiple internal standards for quantification. An interactive web tool should be developed for easy access to the generated data.

Biodiversity of Vitis vinifera endophytes in conventional and biodynamic vineyard

Summary: Plants have developed resistance mechanisms to cope with biotic and abiotic stress, including the presence of microbial endophytes that can promote growth and resilience. This study assessed the diversity and plant growth-promoting properties of bacterial endophytes in two vineyards with different agricultural management approaches in the Czech Republic. A total of 470 bacterial endophytes were isolated, with Bacillus being the dominant genus in both vineyards. The ability of endophytes to produce indole-3-acetic acid (IAA) and ACC deaminase varied between vineyards, with higher antioxidant activity in the conventional vineyard.

POPs and PAHs in fish oil-based food supplements at the Czech market

Summary: The study aimed to evaluate the concentrations of persistent organic pollutants (POPs) and polycyclic aromatic hydrocarbons (PAHs) in 44 fish oil-based food supplements, estimate the daily intake by consumers, and assess the authenticity of the oil samples. The concentrations of SPCBs, OCPs, SPBDEs, and SPAHs found in the samples varied within certain ranges. The results also revealed that some samples labeled as fish oil were likely made from cheaper cod liver oil and had higher levels of halogenated POPs compared to supplements made from fish oil.

Summary: Polycyclic aromatic hydrocarbons (PAHs) were included in the European Human Biomonitoring Initiative (HBM4EU) for harmonization and advancement of human biomonitoring in Europe. A specific QA/QC program using ICIs and EQUASs was developed to ensure comparability and accuracy of analytical laboratories. The study found that 86% of participants achieved satisfactory results in determining 13 PAH metabolites in urine, although low limits of quantification were required. The HBM4EU QA/QC program established an international network of laboratories for comparable analysis of urinary PAH biomarkers.

A Simultaneous Determination of Benzophenone and Camphor UV-Filters, Together with Metabolites of Polycyclic Aromatic Hydrocarbons, in Human Urine

Summary: By monitoring biomarkers in urine, the study assessed personal exposure to selected chemical substances. The results showed the presence of benzophenone and camphor UV-filters, as well as metabolites of polycyclic aromatic hydrocarbons (OH-PAHs), in urine samples collected from mothers and new-borns. The concentrations of these compounds varied across regions and between different countries.

Short-Term Stability of Serum and Liver Extracts for Untargeted Metabolomics and Lipidomics

Summary: Thermal reactions can significantly alter the metabolomic and lipidomic content of biofluids and tissues during storage. In this study, the stability of polar metabolites and complex lipids in dry human serum and mouse liver extracts was investigated over a three-day period under various temperature conditions. Results showed that storing dry extracts at -24 degrees C provided comparable results to -80 degrees C, but increasing the storage temperatures led to significant changes in oxidized triacylglycerols, phospholipids, and fatty acids within three days. Polar metabolites were mainly affected at storage temperatures of +23 degrees C and +30 degrees C.

Pesticide residues in fresh and processed edible mushrooms from Czech markets

Summary: This study investigated pesticide residues in 49 edible mushrooms, finding 21 pesticide residues including prochloraz and carbendazim. The fate of 13 pesticides during mushroom processing was also studied, showing that most pesticide residues remained stable during processing.

Summary: This study investigated the impact of anticoagulants on the molecular characteristics of human BMSCs during isolation and cultivation. The results showed that the type of anticoagulant did not significantly affect the molecular characteristics and cellular composition of BMSCs, while in vitro cultivation caused significant changes in the transcriptomics of BMSCs.
