Summary: Comprehensive two-dimensional gas chromatography with time-of-flight mass spectrometry (GC x GC-TOFMS) is a powerful analytical platform for chemical investigations of complex biological samples. This study optimized a data processing strategy to compensate for 2D pattern misalignments and detector response fluctuations in saliva samples analyzed over 2 years. The strategy was trained on two batches of samples and achieved reliable peak detection and cross-alignment using a combination of untargeted and targeted pattern recognition algorithms and supervised operation with affine functions. Z-score normalization of the mass spectral total useful signal (MSTUS) was found to be the most promising strategy for cross-comparative analysis and data fusion.

Summary: In this study, a comprehensive two-dimensional countercurrent chromatography (CCC) combined with gas chromatography (GC) was used to characterize the chemical constituents of Artemisia argyi essential oil and evaluate the orthogonality of the 2D chromatographic system. A solvent system composed of n-hexane/acetonitrile/methanol (2:2:1, v/v/v) was selected for the first dimensional separation and all CCC fractions were analyzed by GC. A 2D contour plot was created to visualize the chemical compositions. A total of 1392 peaks were obtained through the comprehensive 2D CCC x GC separation and high spatial coverage and low linear correlation coefficient were achieved. Moreover, all compounds were identified by GC-MS. The obtained 2D contour plot could be divided into six zones representing different component groups, which could be used to identify and predict the structure of unknown compounds in Artemisia argyi essential oil for comprehensive characterization.

Summary: This study conducted a comprehensive metabolomic and bioactivity evaluation of different plant parts from Garcinia yunnanensis and Garcinia xanthochymus. Differences in metabolite profiles and bioactivity were observed among different plant parts. The seeds and latex of both species exhibited excellent cytotoxic and antibacterial activities, while the roots of G. xanthochymus and the arils of G. yunnanensis showed strong anti-inflammatory effects.
