Summary: Developing LC-MS/MS methods for detecting native amino acids can be challenging, leading many scientists to derivatise amino acids to improve sensitivity. Derivatisation offers benefits such as reduced signal-to-noise ratios and more efficient ionisation, but it can also be costly and time-consuming. This study aimed to enhance the detection sensitivity of 20 protein amino acids using HILIC-MS/MS.

Summary: The consumption of blue-green algae (BGA) dietary supplements has increased due to their health benefits, but the production of cyanotoxins by cyanobacteria poses serious health risks. This study proposes the use of hydrophilic interaction liquid chromatography coupled with tandem mass spectrometry (HILIC-MS/MS) to detect cyanotoxins in BGA dietary supplements. The method successfully separated and quantified target toxins, including microcystin-leucine-arginine (MC-LR), microcystin-arginine-arginine (MC-RR), nodularin, anatoxin-a, and three non-protein amino acids. The analysis of different commercial supplements revealed the presence of DAB, AEG, and MCs in some samples, emphasizing the importance of monitoring these substances for safe consumption.

Summary: RNA modifications play essential roles in biological activities and are associated with human diseases, including cancers. This study established a method to quantify six nucleosides in urine and plasma samples, and found altered concentrations of certain nucleosides in breast cancer patients. Furthermore, the ratios of methylated adenosine nucleosides to adenosine in plasma were better at distinguishing breast cancer patients from healthy controls.

Summary: Accumulating evidence suggests that DNA methylation, hydroxymethylation, and m6dA play important regulatory roles in tumorigenesis and their aberrations are closely associated with cancer. This study developed a method for accurate quantification of these DNA modifications in human tumor tissues and revealed significant changes in m6dA, 5-mdC, and 5-hmdC levels in different types of cancer.

Summary: The importance of lipidomics research in understanding various diseases, including metabolism, cancer, and the recent COVID-19 pandemic, has led to the development of a targeted HILIC-MS/MS method that allows for comprehensive analysis of lipid species. This method overcomes the challenges posed by the diverse structures and properties of lipids in biological systems, and provides accurate quantitation of lipid concentrations at the fatty acyl chain level. The applicability of this method has been demonstrated through the discovery of differential lipid features related to COVID-19 severity, highlighting its potential for future investigations of the lipidome in different disease contexts.

Summary: Scopolamine is a versatile drug used for various purposes, but overdose can lead to diminished consciousness. The concentration of scopolamine in serum can explain sudden clinical presentations, while its presence in urine can be detected up to 48 hours. Therefore, screening for scopolamine and other toxins is crucial in identifying potentially life-threatening causes.
