Improved procedure for protein binder analysis in mural painting by LC-ESI/Q-q-TOF mass spectrometry: detection of different milk species by casein proteotypic peptides

Diagnostic techniques applied to the field of cultural heritage represent a very important aspect of scientific investigation. Recently, proteomic approaches based on mass spectrometry coupled with traditional spectroscopic methods have been used for painting analysis, generating promising results for binder's protein identification. In the present work, an improved procedure based on LC-ESI/Q-q-TOF tandem mass spectrometry for the identification of protein binders has been developed for the molecular characterization of samples from an early-twentieth-century mural painting from the St. Dimitar Cathedral in Vidin, Bulgaria. The proteomic investigation has led to the identification of both egg white and egg yolk proteins, according to traditional old recipes for tempera paintings. In addition, beyond the egg components, the presence of caseins was also revealed, thus suggesting the use of milk as binding medium, fixative or stabilising agent. Furthermore, for the first time, the capability to discriminate the milk origin on the basis of alpha casein proteotypic peptides is reported, that are diagnostic for a given species, thus opening interesting perspectives in art and archaeological fields.

Understanding the molecular response of P. digitatum to biological and chemical antifungal agents can contribute to the development of safer and more effective non-chemical control methods.

An Updated Review of Bioactive Peptides from Mushrooms in a Well-Defined Molecular Weight Range

Summary: This article reports the current status of bioactive peptides isolated and characterized from mushrooms in the past 20 years. It summarizes the classification, biological activities, isolation principles, and sources of these peptides. It was found that some peptides were misclassified, with certain fungal proteins being mistakenly categorized as peptides. Furthermore, this review highlights the various biological activities exhibited by these peptides.

Ageritin-The Ribotoxin-like Protein from Poplar Mushroom (Cyclocybe aegerita) Sensitizes Primary Glioblastoma Cells to Conventional Temozolomide Chemotherapy

Summary: In this study, Ageritin, the prototype of the ribotoxin-like protein family, was found to inhibit the growth of primary human glioblastoma cell lines and enhance sensitivity to the chemotherapy drug temozolomide. It was also observed to cause genotoxicity through the formation of micronuclei, damaging DNA and downregulating the MGMT DNA repair protein. These findings suggest that Ageritin could be a potential innovative treatment for glioblastoma.

Myoglobin from Atlantic and Tinker mackerels: Purification, characterization and its possible use as a molecular marker

Summary: In this study, the characterization, amino acid sequence determination, and functional comparison of myoglobins (Mbs) from Atlantic mackerel and Tinker mackerel were reported. The results showed differences in autoxidation rate and pseudoperoxidase activity between the two Mbs, which were supported by amino acid sequence and structural analysis. The phylogenetic analysis confirmed that Atlantic mackerel and Tinker mackerel are distinct species. Additionally, an analytical method was developed to distinguish between the two mackerel species.

Tailoring the Structure of Cell Penetrating DNA and RNA Binding Nucleopeptides

Summary: Synthetic nucleic acid interactors have exciting biotechnological and potential therapeutic applications. This study focuses on the synthesis of homo-thymine nucleopeptides and investigates the influence of underivatized amino acid residues and the distance between the nucleobase and peptide backbone on nucleic acid recognition. The results show that two of the nucleopeptides can efficiently bind DNA and RNA targets and cross membranes.

Summary: This study aimed to improve the therapeutic action of cetuximab by conjugating it with the RIP quinoin isolated from quinoa seeds. The novel quinoin cetuximab immunoconjugate showed significant dose-dependent cytotoxicity against cetuximab-resistant cells, suggesting its potential as a candidate for targeted therapy in the treatment of cetuximab-resistant metastatic colorectal cancer.

Structural and functional characterization of the cytotoxic protein ledodin, an atypical ribosome-inactivating protein from shiitake mushroom (Lentinula edodes)

Summary: We have isolated and purified a cytotoxic protein called Ledodin from shiitake mushrooms, which consists of a 197 amino acid chain. Ledodin showed N-glycosylase activity on mammalian 28S rRNA and inhibited protein synthesis, but had no effect on insect, fungal, and bacterial ribosomes. It is believed to have a catalytic mechanism similar to that of DNA glycosylases and plant ribosome-inactivating proteins. Ledodin could be the first representative of a new family of enzymes widely distributed among basidiomycetes. These proteins are of interest due to their potential toxicity in edible mushrooms and their applications in medicine and biotechnology.

A robust nanoLC high-resolution mass spectrometry methodology for the comprehensive profiling of lactic acid bacteria in milk kefir

Summary: Due to the increasing consumer attention to functional foods with probiotics, the market for fermented milk beverage Kefir is growing. There is a demand for new methods to certify the authenticity of Kefir products. In this study, an innovative nanoLC-ESI-MS/MS-based methodology was developed to profile lactic acid bacteria in Kefir products and grains at different levels. This methodology enables the authentication of probiotic starters and provides insights into the composition of lactic acid bacteria in unlabelled Kefir, highlighting the potential of LC-MS/MS as a sensitive tool for assessing the authenticity of fermented foods.

Antifungal Activity of Ageritin, a Ribotoxin-like Protein from Cyclocybe aegerita Edible Mushroom, against Phytopathogenic Fungi

Summary: Ageritin from poplar mushrooms is an antifungal enzyme that inhibits the growth of various fungi affecting crops. It specifically hydrolyzes a phosphodiester bond in the large rRNA, blocking protein synthesis. The antifungal activity of ageritin was demonstrated against Botrytis cinerea, Colletotrichum truncatum, and Alternaria alternata, while no effect was observed on Trichoderma harzianum. In vitro studies revealed that ageritin can damage fungal ribosomes, and further experiments in liquid medium confirmed its concentration-dependent antifungal effect on B. cinerea and T. harzianum.

Nutritional, metabolic and genetic profiling of 'Cerato' and 'Curniciello' bean landraces from Caserta, Southern Italy

Summary: Italy is rich in uncharacterized common bean landraces. This study evaluated the composition and profiles of 'Cerato' and 'Curniciello' dry beans cultivated in Caserta, Southern Italy, and compared them with other local dry beans. 'Cerato' beans had lower levels of proteins, lipids, and amino acids compared to 'Curniciello' beans. Fatty acid analysis showed that linoleic acid, alpha-linolenic acid, and oleic acid were the most abundant in both beans. The inhibitory activities of enzymes related to protein and carbohydrate intake were also investigated. AFLP analysis provided useful information for authentication and traceability. Overall, this study contributes to the preservation of local culinary traditions and Italian biodiversity.
