Alternatives for sample pre-treatment and HPLC determination of Ochratoxin A in red wine using fluorescence detection

Ochratoxin A is a mycotoxin widely studied due to its nephrotoxic, immunotoxic, teratogenic and carcinogenic effects. The European Commission has fixed maximum limits for Ochratoxin A in wines and in other foods. In order to determine Ochratoxin A levels in red wine, the present paper contrasts and discusses the results of a systematic study of analytical parameters for sample pre-treatment using different immunoaffinity cartridges as well as C-18 cartridges with three solvent combinations. The direct injection of wine into two types of C-18 chromatographic columns (conventional packed column and monolithic column) is evaluated as screening method. In all cases, the analysis was carried out using HPLC with fluorescence detection. The results show statistical differences when 3 types of immunoaffinity columns were used, while higher recoveries were obtained for C-18 cartridges using acetonitrile as extraction solvent. Repeatability and accuracy of immunoaffinity and C-18 sample pre-treatment were statistically comparable (alpha = 0.05). Their sensitivity was also comparable, although more favorable detection limits were obtained using the immunoaffinity treatment (0.01 mu g L-1) in comparison with C-18 treatment (0.09 mu g L-1). Considering the maximal allowed concentration of Ochratoxin A in wine (2.00 mu g L-1), both methods are suitable for its determination in wine. Both methods were applied to determine this toxin in 154 wine samples, and the quantitative results demonstrated statistic comparability (alpha = 0.05). These results were also confirmed from the qualitative point of view using a CC-MS method. To find an easy screening method, based on a recent publication, a monolithic HPLC column and 2 conventional packed columns were tested for Ochratoxin A determination in real wine samples by direct injection, without previous clean-up. The results show that this procedure is not useful at the concentration levels usually found in wine and although shorter time is required when using the monolithic columns even with the chromatographic analysis. Finally, based on the results, it was concluded that the combination of C-18 cartridges with conventional particle packed columns and HPLC-FLD is the most appropriate alternative for Ochratoxin A analysis in wine. Summary: Grape canes, known as vine shoots, contain high levels of phenolic compounds that offer a range of health benefits. Extracts from grape canes obtained using a pilot-plant reactor were found to have a different phenolic profile compared to previous laboratory-scale studies. The extracts showed high antioxidant capacity and may potentially be used in the development of novel nutraceutical and pharmaceutical products, increasing the value and sustainability of these residues.

Summary: The study encapsulated grape cane pilot-plant extract in hydroxypropyl beta-cyclodextrin using spray-drying technique, resulting in improved stability and solubility of phenolic compounds with an encapsulation efficiency of 80.5%. The inclusion complex contained 42 compounds with high antioxidant capacity. This formulation has potential applications in water-soluble food and pharmaceutical products.

Metabolic profile and antioxidant capacity of five Berberis leaves species: A comprehensive study to determine their potential as natural food or ingredient

Summary: A comprehensive study on bioactive compounds in the leaves of main Berberis species growing in Chile was conducted. 117 compounds including phenolic acids, alkaloids, flavonols, and other compounds were identified. Hydroxycinnamic acid derivatives and quercetin derivatives were found to be the main compounds. Fatty acid profile and antioxidant capacity also varied among different species. These findings provide insights for novel approaches in utilizing these leaves as natural food or ingredient.

Polydopamine coated hypodermic needles as a microextraction device for the determination of tricyclic antidepressants in oral fluid by direct infusion MS/MS

Summary: In-needle microextraction using hypodermic needles coated with polydopamine was successfully applied to isolate six tricyclic antidepressants from oral fluid samples, achieving efficient preconcentration. Through in-depth study of various variables, optimal extraction conditions and calibration models were established, showing high precision and accuracy of the method.

Native top-down mass spectrometry for monitoring the rapid chymotrypsin catalyzed hydrolysis reaction

Summary: Enzymes play a crucial role as biological catalysts in accelerating biochemical reactions in living organisms. This study proposes a convenient method for monitoring enzymatic catalytic processes using native top-down mass spectrometry. By exploring the heterogeneity of the chymotrypsin sample and using tandem mass spectrometry, the researchers were able to monitor covalent and noncovalent enzymatic complexes, substrates, and products during catalysis. The results demonstrate that this method has the potential to be a promising tool for characterizing biocatalysts.

Carbon fiber-based multichannel solid-contact potentiometric ion sensors for real-time sweat electrolyte monitoring

Summary: This study presents a method for constructing wearable sweat electrolyte sensors using carbon fiber-based solid-contact ion-selective electrodes (SC-ISEs). By using carbon fibers extracted from commercial cloth as electrode material, the cost and reproducibility issues of flexible SC-ISEs were addressed. The results showed that the carbon fiber-based SC-ISEs exhibited reversible voltammetric and stable impedance performances, and had high reproducibility of standard potentials between normal and bending states.

Imaging peroxynitrite in endoplasmic reticulum stress and acute lung injury with a near-infrared fluorescent probe

Summary: A near-infrared fluorescent probe has been developed to accurately measure the levels of peroxynitrite (ONOO-) in the endoplasmic reticulum (ER) in acute lung injury (ALI). The probe demonstrated rapid response, high selectivity, good sensitivity, and enhanced fluorescence intensity in response to ONOO-. It was successfully used to detect changes in ONOO- levels and showed significant increases in an ALI cell model and an ALI mouse model.

Miniaturized ascorbic acid assay platform based on point discharge atomic emission spectrometry coupling with gold filament enrichment

Summary: In this work, a portable analytical system based on point discharge chemical vapor generation atomic emission spectrometry (PD-CVG-AES) coupling with gold filament enrichment was designed. The highly sensitive analysis of Hg2+ indirectly realized the detection of ascorbic acid (AA). The measurement is based on the fact that Ag+ can decrease the concentration of Hg2+ by forming Ag-Hg amalgam in the presence of the reductant SnCl2, while AA can pre-reduce Ag+ to Ag0, leading to the generation of silver nanoparticles (Ag NPs). The developed novel analytical strategy broadens the application of microplasma-based AES and offers a higher level of sensitivity compared to current AA detection techniques.

Time-gated luminescent probes for lysosomal singlet oxygen: Synthesis, characterizations and bioimaging applications

A novel polymer-based probe for fluorescently ratiometric sensing of hydrogen sulfide with multiple applications

Summary: A novel polymer-based probe P1 was successfully synthesized for fluorescently ratiometric sensing of H2S with high selectivity and sensitivity. A smartphone sensing platform was constructed to conduct visual quantitative detection of H2S. P1 can be employed in evaluating the level fluctuations of H2S in living cells, testing water samples/wine samples, and monitoring food freshness.

Split aptamer-based sandwich-type ratiometric biosensor for dual-modal photoelectrochemical and electrochemical detection of 17β-estradiol

Summary: The study developed a split aptamer-based sandwich-type ratiometric biosensor for the detection of 17 beta-estradiol (E2) using photoelectrochemical and electrochemical assays. The biosensor utilized split aptamer fragments to recognize E2 and trigger a hybridization chain reaction, resulting in the production of double-stranded DNA labeled with CdTe quantum dots. This DNA complex was able to sensitize CdTe quantum dots and generate response signals for E2 detection. The developed biosensor demonstrated high sensitivity and accuracy with two linear ranges and low detection limits.
