Advances in optical biopsy - correlation of malignancy and cell density of primary brain tumors using Raman microspectroscopic imaging

Raman spectroscopy is a promising tool towards biopsy under vision as it provides label-free image contrast based on intrinsic vibrational spectroscopic fingerprints of the specimen. The current study applied the spectral unmixing algorithm vertex component analysis (VCA) to probe cell density and cell nuclei in Raman images of primary brain tumor tissue sections. Six Raman images were collected at 785 nm excitation that consisted of 61 by 61 spectra at a step size of 2 micrometers. After data acquisition the samples were stained with hematoxylin and eosin for comparison. VCA abundance plots coincided well with histopathological findings. Raman spectra of high grade tumor cells were found to contain more intense spectral contributions of nucleic acids than those of low grade tumor cells. Similarly, VCA endmember signatures of Raman images from high grade gliomas showed increased nucleic acid bands. Further abundance plots and endmember spectra were assigned to tissue containing proteins and lipids, and cholesterol microcrystals. Since no sample preparation is required, an important advantage of the Raman imaging methodology is that all tissue components can be observed - even those that may be lost in sample staining steps. The results demonstrate how morphology and chemical composition obtained by Raman imaging correlate with histopathology and provide complementary, diagnostically relevant information at the cellular level.

Surface-Enhanced Raman Spectroscopy to Characterize Different Fractions of Extracellular Vesicles from Control and Prostate Cancer Patients

Summary: Extracellular vesicles (EVs) ranging in size from 60 to 800 nm were isolated from plasma of control and prostate cancer patients using differential centrifugation and ultrafiltration techniques. Spontaneous Raman and surface-enhanced Raman spectroscopy (SERS) were employed to identify fractions that best discriminate between control and cancer patients, with results showing that EV12 and EV120 fractions provide the most effective separation.

Structural and Biochemical Changes in Pericardium upon Genipin Cross-Linking Investigated Using Nondestructive and Label-Free Imaging Techniques

Summary: This study investigated the biochemical, structural, and biomechanical effects of genipin (GE) cross-linking on equine pericardium (EP) using optical imaging techniques and atomic force microscopy (AFM). Results showed that GE cross-linking led to a decrease in second-harmonic signal and an increase in autofluorescence, indicating changes in biochemical and structural properties. Additionally, cross-linking increased the Young's modulus of EP.

Sensitive determination of uranium using ß-cyclodextrin modified graphene oxide and X-ray fluorescence techniques: EDXRF and TXRF

Summary: In this study, ss-cyclodextrin/graphene oxide (GO-ss-CD) was used as an effective adsorbent for dispersive micro-solid phase extraction (DMSPE) of uranyl ions (UO22+) from water samples. The adsorption capacity and resistance to high ionic strength make it a potential method for preconcentration of UO22+ ions. The accuracy of the method was verified by analyzing certified reference material and spiked water samples.

Opportunities of optical and spectral technologies in intraoperative histopathology

Summary: Modern optical and spectral technologies have the potential to characterize tissues at a molecular level, enabling the identification of pathological tissues and the grading and staging of tumors without the need for labeling. The current tools for intraoperative tumor assessment, as well as the requirements for tissue visualization from a medical and optical perspective, are described. Approaches utilizing optical and spectral techniques for ex vivo and in vivo monitoring are introduced, and proof-of-concept studies utilizing linear and nonlinear spectroscopy and imaging modalities are presented. The use of modern artificial intelligence and deep learning concepts to analyze spectroscopic and imaging datasets has contributed to the advancements in each technique. Finally, the article provides an outlook on the opportunities and prospects of clinical translation.

Raman Spectroscopic Imaging of Human Bladder Resectates towards Intraoperative Cancer Assessment

Summary: Raman spectroscopy allows for label-free assessment of bladder tissue in intraoperative applications. In a retrospective study, Raman microspectroscopic images were used to identify different components of bladder tissue, including normal bladder, bladder cancer, necrosis, epithelium, and lipid inclusions. Additionally, foreign materials such as microplastic particles and pigments were detected in the specimens. This study demonstrates the potential of Raman spectroscopy for tumor detection and material identification in complex matrices.

Summary: This study demonstrates how the O-PTIR technique can overcome the limitations of direct mid-infrared spectroscopic imaging of single cells in an aqueous buffer. By using an indirect detection principle, the technique achieves high-resolution discrete wavenumber imaging and fingerprint spectroscopy of cultivated cells. The study also shows the potential applications of O-PTIR in high-content screening and subcellular analysis.
