Confocal Raman microspectroscopy as an analytical tool to assess the mitochondrial status in human spermatozoa

Confocal Raman microspectroscopy was used to investigate human sperm cells. Raman mapping with a 532 nm excitation laser allowed to unambiguously characterize the nucleus, the neck, and, in particular, the mitochondria-rich middle piece of a human sperm cell. The effect of ultraviolet radiation on different organelles of the sperm was quantified by localized spectral Raman signatures obtained within milliseconds. Chemical changes within the sub-cellular structure of the sperm cells were recorded as a function of ultraviolet light exposure time, showing the proof-of-principle that Raman microspectroscopy can be a fast diagnostic method for detecting the mitochondrial and motility status of human spermatozoa.

Accelerated Deep Reinforcement Learning for Fast Feedback of Beam Dynamics at KARA

Summary: Coherent synchrotron radiation (CSR) is generated when the electron bunch length is in the order of the magnitude of the wavelength of the emitted radiation. To stabilize the CSR emission, a real-time feedback control loop based on reinforcement learning (RL) is proposed. The hardware implementation of the RL controller shows similar training performance to the software solution, but outperforms the Keras-RL implementation by an order of magnitude, indicating its potential as an essential platform for developing intelligent CSR control systems.

pH effects on the molecular structure and charging state of β-Escin biosurfactants at the air-water interface

Summary: Saponins like beta-escin have high surface activity and unique surface rheology, making them interesting for applications in soft matter colloids and interfaces. Using vibrational sum-frequency generation (SFG), beta-escin adsorption layers at the air-water interface were studied, showing changes in O-H intensity with pH due to interfacial charging state and deprotonation of carboxylic acid. The SFG spectra also revealed drastic changes in C-H bands from interfacial beta-escin, related to differences in molecular orientation.

True Origin of Amide I Shifts Observed in Protein Spectra Obtained with Sum Frequency Generation Spectroscopy

Summary: Accurate determination of protein structure at interfaces is critical for understanding protein interactions, and VSFG spectroscopy is often used for this purpose. However, our study shows that the peaks shifts observed in conventional VSFG spectra upon changing the solution pH are actually due to nonresonant contributions. Therefore, HD-VSFG measurements are necessary for drawing unambiguous conclusions about structural changes in biomolecules.

In-line measurement of fiber mass fraction using Terahertz spectroscopy for a function-oriented quality assurance of glass fiber sheet molding compound

Summary: This study extends a non-destructive measurement model for local fiber mass fraction using Terahertz spectroscopy, suitable for multiple specimen thicknesses. A thorough analysis of measurement uncertainty shows a significant reduction compared to manufacturing process uncertainty, with short measurement times suitable for in-line applications. Integrating the measurement results into a parametrized finite element model reduces the error between physical experiments and simulations.

Lichen species across Alaska produce highly active and stable ice nucleators

Summary: Forty years ago, lichens were found to possess extraordinary ice nucleation abilities, allowing them to thrive in freezing environments. The ice nucleators in lichens have the potential to impact cloud glaciation and atmospheric processes in the warming Arctic. This study investigates the ice nucleation capabilities of lichens collected from different ecosystems in Alaska, and finds widespread ice nucleating activity, particularly in the coastal rainforest of southeast Alaska. These findings suggest that the ice nucleation activity is an essential trait for the survival of lichens in cold environments and can influence local atmospheric processes.
