The pathoconnectivity network analysis of the insular cortex: A morphometric fingerprinting

Andrea Nani, Jordi Manuello, Lorenzo Mancuso, Donato Liloia, Tommaso Costa, Alessandro Vercelli, Sergio Duca, Franco Cauda

Summary: Brain disorders tend to affect the insular cortex, showing patterns of co-alteration with a dense network including predominantly cortical regions. Frontal lobe regions are more involved than occipital lobe, and co-alteration and co-activation patterns overlap significantly. This suggests alterations caused by brain disorders follow a network architecture logic, with brain hubs at the center of co-altered areas.

Critical illness neuro-myopathy (CINM) and focal amyotrophy in intensive care unit (ICU) patients with SARS-CoV-2: a case series

Nicola Alessandro Nasuelli, Roberto Pettinaroli, Laura Godi, Claudio Savoini, Fabiola De Marchi, Letizia Mazzini, Federico Crimaldi, Aline Pagni, Cleta Patrizia Pompa, Davide Colombo

Summary: The study identified four patients with specific phenotype in the ICU, characterized by focal hypotrophies likely caused by the prone position. The team intends to initiate early treatment protocols with electrical stimulation to evaluate its effectiveness in preventing critical illness and focal hypotrophies in ICU SARS-CoV-2 patients, aiming to improve the likelihood of returning to pre-infection functional status.

Summary: This study analyzed the toxicity and outcome predictors in Ewing sarcoma patients with lung metastases treated with high-dose chemotherapy followed by whole-lung irradiation. The results indicated that this combined treatment strategy is feasible and may help in controlling the disease. Further research is needed to explore treatment stratification based on the histological response of the primary tumor.

Identification of Biochemical and Molecular Markers of Early Aging in Childhood Cancer Survivors

Silvia Ravera, Tiziana Vigliarolo, Silvia Bruno, Fabio Morandi, Danilo Marimpietri, Federica Sabatini, Monica Dagnino, Andrea Petretto, Martina Bartolucci, Monica Muraca, Eleonora Biasin, Riccardo Haupt, Marco Zecca, Franca Fagioli, Daniela Cilloni, Marina Podesta, Francesco Frassoni

Summary: This study aimed to identify the mechanisms involved in accelerated aging, increased risk of second tumors, and chronic diseases in childhood cancer survivors due to chemo/radiotherapy. Biochemical and molecular analyses revealed inefficient oxidative phosphorylation, low energy status, and altered metabolism in CCS-MNCs compared with healthy controls, suggesting potential targets for pharmacological interventions to restore mitochondrial function and slow down aging-associated pathologies in CCS.

A Novel Xeno-Free Method to Isolate Human Endometrial Mesenchymal Stromal Cells (E-MSCs) in Good Manufacturing Practice (GMP) Conditions

Stefano Canosa, Katia Mareschi, Elena Marini, Andrea Roberto Carosso, Sara Castiglia, Deborah Rustichelli, Ivana Ferrero, Gianluca Gennarelli, Benedetta Bussolati, Alberto Nocifora, Valentina Asnaghi, Massimiliano Bergallo, Ciro Isidoro, Chiara Benedetto, Alberto Revelli, Franca Fagioli

Summary: The study compared two different endometrial sampling methods and validated a novel xeno-free method for culturing human E-MSCs. Results showed that VABRA endometrial sampling could be an alternative to surgical curettage, providing efficient isolation and culture of E-MSCs in Good Manufacturing Practice compliance conditions.

A New Human Platelet Lysate for Mesenchymal Stem Cell Production Compliant with Good Manufacturing Practice Conditions Preserves the Chemical Characteristics and Biological Activity of Lyo-Secretome Isolated by Ultrafiltration

Katia Mareschi, Alessia Giovanna Santa Banche Niclot, Elena Marini, Elia Bari, Luciana Labanca, Graziella Lucania, Ivana Ferrero, Sara Perteghella, Maria Luisa Torre, Franca Fagioli

Summary: This study modified the type of culture supplement used in the production of freeze-dried mesenchymal stem cell (MSC)-secretome (lyo-secretome), and found that changing the supplement type only resulted in minimal changes in product composition without affecting potency, validating the new process according to GMP standards.

Summary: This study demonstrates that the use of HPL-S is an effective alternative for MSC production under GMP conditions. The protein and growth factor content in HPL-S is comparable to that of HPL-E. Furthermore, MSCs derived from HPL-S show lower HLA-DR expression in immunophenotypic analysis.

Work Placement and Job Satisfaction in Long-Term Childhood Cancer Survivors: The Impact of Late Effects

Margherita Dionisi-Vici, Alessandro Godono, Anna Castiglione, Filippo Gatti, Nicoletta Fortunati, Marco Clari, Alessio Conti, Giulia Zucchetti, Eleonora Biasin, Antonella Varetto, Enrico Pira, Franca Fagioli, Enrico Brignardello, Francesco Felicetti

Summary: This study aims to describe the occupational status of long-term childhood cancer survivors (CCS) and evaluate the association between late effects and unemployment and job satisfaction. The results show that severe late effects have a negative impact on occupation and worsen the perception of satisfaction with the economic situation of unemployed CCS. Routine follow-up care for CCS should include monitoring their socioeconomic development and providing interventions to help them obtain suitable jobs.

No Child Should Be Left Behind by COVID-19: A Report about the COVID-19 Pandemic Experience in Children and Adolescents with Acute or Chronic Disease Treated at a Pediatric Referral Hospital in Italy

Giulia Zucchetti, Sabrina Ciappina, Cristiana Risso, Alice Malabaila, Sara Racalbuto, Elena Longo, Margherita Dionisi Vici, Marina Bertolotti, Paola Quarello, Franca Fagioli

Summary: This study investigated the feelings of children and adolescents with acute or chronic illness towards the COVID-19 pandemic in Italy, and compared their experiences with children without illness. The results showed that participants, regardless of their health condition, felt fear of the virus and potential infection for themselves and their families, but experienced less interference with daily functioning. The fragile group demonstrated greater resilience towards the pandemic situation compared to the low-risk group, and some differences were found within the fragile group based on the type of illness.
