Mesenchymal Stem Cells: Therapeutic Mechanisms for Stroke

Summary: Due to the aging population, stroke has become more prevalent, leading to increased socioeconomic burden. Stem cell transplantation, particularly using mesenchymal stem cells (MSCs), has shown promise in treating stroke by attenuating inflammation, promoting therapeutic effects, inducing blood vessel formation, promoting neurogenesis, reducing infarct volume, and replacing damaged cells. This review discusses the mechanisms of MSCs and their secreted extracellular vehicles (EVs) in stroke treatment.

However, during the inflammatory process, MSCs become impaired and lose their immunomodulatory activity. Despite challenges and risks, MSCs show promise in the treatment of severe diseases.

Summary: Type 1 diabetes mellitus leads to degeneration of organelles in adipose-derived stem cells, with the main characteristic being prominent enlarged spherical vesicles. The exosomes of T1DM ADSCs are found to be enlarged, reduced in number, and increased in the percentage positive for tetraspanin CD9, providing insight into the characteristics of stem cells in patients with type 1 diabetes mellitus.

Hydrogels for Large-Scale Expansion of Stem Cells

Summary: Water-based hydrogels have shown significant progress in regulating stem cell proliferation and differentiation by mimicking the native microenvironment, including bioavailability, mechanical strength, and porosity. Tailoring hydrogel properties to suit different stem cell types can effectively enhance their proliferation and expansion for various applications. Future research directions aim to improve current in vitro stem cell expansion systems by constructing hydrogels that closely resemble the microenvironment of stem cells in vivo.

Long-term survival benefits of intrathecal autologous bone marrow-derived mesenchymal stem cells (Neuronata-R®: lenzumestrocel) treatment in ALS: Propensity-score-matched control, surveillance study

Summary: This study aimed to investigate the long-term survival benefits of intrathecal lenzumestrocel treatment. The results showed that the survival probability was significantly higher in the BM-MSC group compared to the external control group. Multivariate Cox regression analysis indicated a significantly lower hazard ratio for death in the BM-MSC group, and multiple injections were found to be more effective. Additionally, no serious adverse drug reactions were observed during the safety assessment lasting a year after the first administration.

Summary: In this study, a new microcarrier with hyaluronic acid (HA) as the core material was developed for scalable serum-free suspension culture of hESC-derived MSCs. The optimal microcarriers, with a coating collagen concentration of 100 μg/mL or concave-structured surface (cHAMCs), allowed for the expansion of IMRCs within six days with a high yield. The harvested cells showed high viability, immunomodulatory and regenerative therapeutic promise, as well as increased secretion of extracellular matrix (ECM).

Mesenchymal stem cells-derived exosomes for drug delivery

Summary: Exosomes are extracellular vesicles secreted by various cells, mainly composed of lipid bilayers without organelles. Researchers have increasingly focused on using exosomes for drug delivery, especially MSC-EXOs, due to their immune properties, tumor-homing properties, and elastic properties. This review article summarizes the characteristics of MSC-EXOs and introduces production, purification, and drug loading methods to address existing problems and provide suggestions for future studies.

Cell therapy in ALS: An update on preclinical and clinical studies

Summary: Amyotrophic lateral sclerosis (ALS) is a progressive neurodegenerative disease characterized by loss of motor neurons and neuromuscular impairment. Stem cell therapy shows promise in protecting motor units through various mechanisms. However, more coordinated effort and research is needed to address challenges in stem cell source selection, mechanism of action, and expected clinical outcomes. This review covers recent advances in stem cell therapies in ALS animal models and ongoing human clinical trials.

After injection into the joint cavity, the microspheres expedite the release of mesenchymal stem cells (MSCs) in the microenvironment of arthritis inflammation, while using kartogenin to promote the differentiation of MSCs into chondrocytes, thereby accelerating articular cartilage regeneration.

The pathoconnectivity network analysis of the insular cortex: A morphometric fingerprinting

Summary: Brain disorders tend to affect the insular cortex, showing patterns of co-alteration with a dense network including predominantly cortical regions. Frontal lobe regions are more involved than occipital lobe, and co-alteration and co-activation patterns overlap significantly. This suggests alterations caused by brain disorders follow a network architecture logic, with brain hubs at the center of co-altered areas.

Summary: This study aimed to identify the mechanisms involved in accelerated aging, increased risk of second tumors, and chronic diseases in childhood cancer survivors due to chemo/radiotherapy. Biochemical and molecular analyses revealed inefficient oxidative phosphorylation, low energy status, and altered metabolism in CCS-MNCs compared with healthy controls, suggesting potential targets for pharmacological interventions to restore mitochondrial function and slow down aging-associated pathologies in CCS.

Summary: This study aims to describe the occupational status of long-term childhood cancer survivors (CCS) and evaluate the association between late effects and unemployment and job satisfaction. The results show that severe late effects have a negative impact on occupation and worsen the perception of satisfaction with the economic situation of unemployed CCS. Routine follow-up care for CCS should include monitoring their socioeconomic development and providing interventions to help them obtain suitable jobs.

No Child Should Be Left Behind by COVID-19: A Report about the COVID-19 Pandemic Experience in Children and Adolescents with Acute or Chronic Disease Treated at a Pediatric Referral Hospital in Italy

Summary: This study investigated the feelings of children and adolescents with acute or chronic illness towards the COVID-19 pandemic in Italy, and compared their experiences with children without illness. The results showed that participants, regardless of their health condition, felt fear of the virus and potential infection for themselves and their families, but experienced less interference with daily functioning. The fragile group demonstrated greater resilience towards the pandemic situation compared to the low-risk group, and some differences were found within the fragile group based on the type of illness.
