Summary: This study developed a cellular model of Bardet-Biedl syndrome (BBS) using induced pluripotent stem cell-derived hypothalamic arcuate-like neurons. The findings suggest that obesity in BBS patients may be related to morphological defects in neurons, impaired insulin signaling, and leptin-mediated p-STAT3 activation. The data provide insights into the mechanisms by which components of the BBSome in central nervous system primary cilia affect energy homeostasis.

Summary: By reviewing studies on the FTO locus in obesity genetics, this article highlights the advances in assessing molecular bases for genetic associations through the use of technological and analytical strategies. The article focuses on extrapolating experimental findings from animal models and cell types to humans, as well as the technical aspects used to identify long-range DNA interactions and their biological relevance. A unifying model is proposed, integrating multiple FTO variants and genes that regulate independent obesogenic pathways at the primary cilium, a cellular antenna for energy balance signaling molecules.
