Summary: The paradigm of mast cells in type 2 diabetes is changing. Research has found that mast cells, once considered harmful inflammatory cells, play important roles in adipose tissue homeostasis. A study measuring the surface expression of four proteins on mast cells in the adipose tissue of obese patients with different stages of diabetes found that type 2 diabetes affects mast cells more severely in omental adipose tissue than in subcutaneous adipose tissue.

Summary: Body tissues can experience temporary hypoxia due to variations in oxygen levels. The hypoxic response is regulated by hypoxia-inducible factor (HIF), which influences cellular metabolism, immune responses, barrier integrity, and microbiota. Recent studies have investigated the response to hypoxia during various infections, but the role of HIF activation in protozoan parasitic infections is still poorly understood. Evidence suggests that protozoa can activate HIF and target genes in the host, affecting their pathogenicity. This review focuses on the hypoxic response to protozoan infections and its impact on host immune responses.

Summary: Hypoxia, a common characteristic of solid tumors, contributes to different aspects of tumor progression and limits the efficacy of immunotherapies. Developing new immunotherapy strategies involving therapeutic targeting of HIF-1 molecules associated with hypoxia may enhance the clinical effectiveness of immunotherapy. Targeting hypoxia presents a potential opportunity to improve the clinical benefit of cancer immunotherapy.

This review summarizes recent findings on the role of hypoxia and HIFs in EwS, suggesting their potential clinical relevance as prognostic markers and therapeutic targets in EwS treatment.

Summary: Hypoxia-inducible factor-1 alpha (HIF-1 alpha) is highly expressed in hypoxic tumors, including hepato-cellular carcinoma (HCC). Another key transcription factor, nuclear factor erythroid 2-related factor 2 (NRF2), is also overactivated in HCC. The interaction between NRF2 and HIF-1 alpha contributes to HCC growth and progression.
