Summary: The Ca2+ modulated pulsatile glucagon and insulin secretions by pancreatic alpha and beta cells play a crucial role in glucose homeostasis. However, the coordination between alpha and beta cells in producing different Ca2+ oscillation patterns is still unclear. This study used a microfluidic device and transgenic mice to record Ca2+ signals from islet alpha and beta cells and observed heterogeneous Ca2+ oscillation patterns intrinsic to each islet. The results showed that after glucose stimulation, the oscillations of alpha and beta cells were globally phase-locked, with a fixed time delay in the activation of alpha cells compared to beta cells and a tunable period in the activation of beta cells. The number of islet alpha cells was also found to be correlated with oscillation frequency. A mathematical model incorporating paracrine interactions was built and quantitatively agreed with the experimental data, highlighting the importance of cell-cell interaction in generating stable but tunable islet oscillation patterns.

Summary: GC-globulin (GC), also known as vitamin D-binding protein, plays a multifunctional role in transporting vitamin 25(OH)D, fatty acids, and scavenging actin. It is highly localized to glucagon-secreting alpha-cells in the pancreatic islets. Loss of GC exacerbates alpha-cell failure during metabolic stress, leading to impaired glucagon secretion and changes in alpha-cell morphology. Restoring GC levels rescues the impairments in glucagon secretion and restores the F-actin cytoskeleton of alpha-cells. Decreased GC levels in alpha-cells of type 2 diabetes donors are associated with changes in alpha-cell mass, morphology, and glucagon expression.

Summary: This study aimed to resolve conflicting findings about GLP-1 levels in different glucose tolerance states among Malay, Chinese, and Indian ethnicities. The results showed that GLP-1 levels were higher in T2DM and pre-DM compared to NGT, suggesting a possible adaptive compensatory response. Among the three ethnic groups, Indians had the highest levels of GLP-1 and insulin resistance, supporting the notion of an adaptive compensatory secretion of GLP-1.

Summary: In this study, the potential effects of alpha-Spinasterol, a phytosterol isolated from Aster pseudoglehnii, on glucose uptake and insulin secretion in skeletal muscle cells and pancreatic beta-cells were investigated. The results showed that alpha-Spinasterol enhanced glucose uptake and insulin secretion without causing any toxic effects on the cells. It also increased the expression of proteins related to glucose metabolism. These findings suggest that alpha-Spinasterol could be a potential candidate for the treatment of type 2 diabetes.
