Summary: Bone remodeling is crucial for maintaining bone quality by removing and replacing damaged and old bone. This process requires tight coordination between bone resorption and bone formation. Besides osteoclasts and osteoblasts, macrophages and other myeloid lineage cells have been found to support bone remodeling. This article discusses the origin and functions of macrophages in the bone microenvironment, including tissue resident macrophages, osteomacs, and newly identified osteomorphs resulting from osteoclast fission. The role of macrophages in inflammatory bone resorption is also touched upon.

Patients with ISM have more vertebral fractures, and women tend to be younger. This study highlights the importance of considering ISM in premenopausal women and men with vertebral fractures even without typical skin involvement.

Summary: Oral bisphosphonates (BPs) are commonly used for the treatment of osteoporosis. This study found that long-term BP treatment can modulate the expression of specific microRNAs (miRNAs), specifically miR-30a-5p, which is negatively correlated with bone formation. In vitro and in vivo experiments demonstrated that miR-30a-5p inhibits osteoblastic differentiation and bone formation. This research provides important insights into the regulatory mechanisms of long-term BP treatment in postmenopausal osteoporosis.

Summary: miRNA is an ideal master regulator for organogenesis due to its ability to simultaneously modulate a set of genes for lineage-specific development. However, the lack of obvious phenotypic defects in most miRNA deletions may be attributed to functional redundancy. Our study identifies MIR27a as a specific miRNA essential for bone homeostasis, as its loss causes severe osteoporosis in mice. MIR27a affects bone resorption by osteoclasts and not bone formation by osteoblasts, and its effects are mediated through the regulation of Squstm1/p62.

When exposed to an SMF, IONPs and SMFs synergistically enhance their respective effects on osteoblasts and osteoclasts.
