Distinct Laminar and Cellular Patterns of GABA Neuron Transcript Expression in Monkey Prefrontal and Visual Cortices

Samuel J. Dienel, Andrew J. Ciesielski, Holly H. Bazmi, Elizabeth A. Profozich, Kenneth N. Fish, David A. Lewis

Summary: The functional output of a cortical region is influenced by its complement of GABA neuron subtypes. Differences in expression patterns of GABA-related transcripts were found between the primate DLPFC and V1, reflecting the diversity in GABA neuron subtypes and gene expression levels per neuron. These findings suggest that specialization of inhibitory neurotransmission across cortical circuits is influenced by variations in the complement of GABA neuron subtypes and gene expression levels per neuron.

Laminar Differences in the Targeting of Dendritic Spines by Cortical Pyramidal Neurons and Interneurons in Human Dorsolateral Prefrontal Cortex

Jill R. Glausier, Dibyadeep Datta, Kenneth N. Fish, Daniel W. Chung, Darlene S. Melchitzky, David A. Lewis

Summary: Activation of specific neural circuits in different layers of the primate dorsolateral prefrontal cortex is crucial for working memory, with recurrent excitation between pyramidal neurons contributing to laminar-specific activity. The density of spines receiving input from cortical pyramidal neurons is higher in the middle layer compared to the deep layer in the human DLPFC, and there are also dendritic spines dually innervated by cortical pyramidal neurons and interneurons. These findings suggest a microcircuit structure in the middle laminar zone that supports the modulation of synaptic information in recurrent excitatory microcircuits.

Vesicular glutamate transporter modulates sex differences in dopamine neuron vulnerability to age-related neurodegeneration

Summary: The study revealed the impact of age and sex differences on dopamine neuron vulnerability, with males exhibiting greater decreases in neuron number and locomotion during aging, and dynamic changes in VGLUT expression in dopamine neurons possibly serving as a compensatory mechanism. Female fruit flies were found to possess higher levels of VGLUT expression in dopamine neurons compared to males, and diminishing VGLUT expression eliminated their greater resilience to neuron loss. Optimal control over VGLUT expression in dopamine neurons was identified as essential for neuron survival and a potential therapeutic target for age- and PD-related neurodegeneration.

VGLUT2 Is a Determinant of Dopamine Neuron Resilience in a Rotenone Model of Dopamine Neurodegeneration

Silas A. Buck, Briana R. De Miranda, Ryan W. Logan, Kenneth N. Fish, J. Timothy Greenamyre, Zachary Freyberg

Summary: The study found that DA neurons expressing VGLUT2 in the VTA/SNc exhibit higher resilience to rotenone-induced neurodegeneration, with an increase in VGLUT2 expression in response to rotenone. Dopaminergic terminals with VGLUT2 expression show greater protection throughout the striatum.

Machine learning sequence prioritization for cell type-specific enhancer design

Alyssa J. Lawler, Easwaran Ramamurthy, Ashley R. Brown, Naomi Shin, Yeonju Kim, Noelle Toong, Irene M. Kaplow, Morgan Wirthlin, Xiaoyu Zhang, BaDoi N. Phan, Grant A. Fox, Kirsten Wade, Jing He, Bilge Esin Ozturk, Leah C. Byrne, William R. Stauffer, Kenneth N. Fish, Andreas R. Pfenning

Summary: Recent discoveries of extreme cellular diversity in the brain necessitate the development of technologies to access specific cell populations within heterogeneous tissue. This study introduces a virus-based strategy called Specific Nuclear-Anchored Independent Labeling (SNAIL) that uses machine learning to identify cell type-specific gene activation features and enables cell labeling and nuclear isolation. The SNAIL approach has been successfully validated for parvalbumin-expressing neurons and shows promise for cell type-specific observation and manipulation in different species and disease models.

Cell type specific cannabinoid CB1 receptor distribution across the human and non-human primate cortex

Shinnyi Chou, Tejis Ranganath, Kenneth N. Fish, David A. Lewis, Robert A. Sweet

Summary: Changes in the cannabinoid CB1 receptor (CB1R) are related to various psychiatric disorders. CB1R participates in regulating the balance between excitatory and inhibitory signals. Previous studies mostly focused on rodents, so it is important to investigate the distribution patterns of CB1R in the human and non-human primate cortex. The findings show that CB1R is present in both excitatory and inhibitory boutons, with higher levels in inhibitory boutons.

Laminar-Specific Alterations in Calbindin-Positive Boutons in the Prefrontal Cortex of Subjects With Schizophrenia

Kenneth N. Fish, Brad R. Rocco, James D. Wilson, David A. Lewis

Summary: This study found that GABA neurons in the prefrontal cortex of patients with schizophrenia are impaired. The decrease in GAD67 messenger RNA levels in these patients affects a subset of calbindin-expressing GABA neurons.

Summary: By studying mouse models with knockdown of the SETD1A and DISC1 genes, it was found that XL synapses can lead to excessive neuronal firing and impair working memory. This finding provides new evidence for understanding the synaptic pathology of psychiatric disorders.
