Summary: Secondary lymphedema is a result of lymphatic insufficiency following surgery or radiation for breast or pelvic cancer. It leads to reduced quality of life, skin ulcers, pain, and infection. Current countermeasures mainly involve physical therapy, but there is a need for a fundamental and effective treatment. Therapeutic lymphangiogenesis shows promise in reconstructing the lymphatic network and recovering lymphatic vessel function, offering a potential cure for lymphedema. This review focuses on the translational research and prospects of therapeutic lymphangiogenesis for lymphedema.

Summary: This study prepared and characterized lipidic nanocapsules loaded with melatonin and demonstrated their efficacy in treating ischemic stroke. The results showed that the nanocapsules had good stability and sustained release ability, and achieved efficient drug permeation across sheep nasal mucosa. The study also found that MEL-LNCs could reduce oxidative stress, decrease inflammatory response, and promote neuronal survival.

LRRC8A has been identified as a potential prognostic biomarker and therapeutic target for PAAD, as it affects the hallmarks of PAAD and is correlated with cell proliferation, cell migration, drug resistance, and immune infiltration. These findings can contribute to the development of new therapeutic strategies for PAAD.

Summary: This study explored the therapeutic efficacy of local steroid injections (LSI) in patients with severe idiopathic granulomatous mastitis (IGM). The results showed that LSI could be considered as the first-line treatment for severe IGM, either as a sole treatment or in combination with oral steroids. The complete response rates did not differ between the LSI group and the combined LSI with oral steroid treatment group, but systemic side effects were lower in the LSI alone group.

Summary: Digital therapeutics (DTx) are software applications embedded within accessible technologies to treat, manage, or prevent pathological conditions. The development of DTx holds great promise for improving patients' lives, but generating therapeutic evidence for DTx presents challenges. By applying clinical pharmacology principles, such as mechanism of action characterization, intervention optimization, and dosing determination, the development of DTx could be enhanced. Our review of DTx studies highlights the importance of integrating clinical pharmacology principles from traditional therapeutics development with considerations from the world of digital solutions.

Summary: CARE-Express is an intervention developed for older adults with cancer to enhance coping and reduce psychological distress. This study evaluated the feasibility and initial efficacy of CARE-Express. The results showed that CARE-Express had positive effects on reducing depression and demoralization, as well as increasing spirituality and faith. The effects attenuated at four months, but small to moderate positive effects remained.

Summary: In recent decades, there have been significant advancements in the diagnosis and treatment of systemic light chain (AL) amyloidosis, leading to improved outcomes and decreased early mortality. However, wide survival gaps still exist among different patient subgroups, with most deaths attributed to disease-related factors such as cardiac failure and sudden unexpected death. AL amyloidosis-unrelated mortality has increased over time and with longer-term survival, highlighting the changing landscape of care standards and the overall optimistic outlook for patients with AL amyloidosis.
