This study showed that high dietary sodium chloride (NaCl) intake increased hydrogen peroxide (H2O2) production by endothelial cells, leading to osteogenic differentiation of vascular smooth muscle cells. These findings revealed a cell-cell communication mechanism in the vascular wall involving H2O2 production by endothelium and its regulation by dietary NaCl intake.

Summary: This study aimed to investigate the effect of a 7-day high-salt diet on the initiation of vascular low-grade inflammation in young healthy individuals and the specific role of oxidative stress. The results showed that short-term high-salt diet increased levels of pro-inflammatory and anti-inflammatory cytokines, as well as oxidative stress. It also affected the frequency of Treg cells, suggesting alterations in immune regulatory mechanisms.

Summary: Pulmonary arterial hypertension (PAH) is a disease characterized by progressive pulmonary vascular remodeling, leading to increased pulmonary artery pressure and right heart dysfunction. There is evidence of cognitive impairment in PAH, but the underlying mechanism and the relationship between cardiopulmonary and cognitive dysfunction in PAH are still unknown. In this study, cognitive evaluations and sub-maximum cardiopulmonary exercise testing were performed on adult subjects with PAH. It was found that 26% of the subjects exhibited a frontal-subcortical syndrome suggestive of vascular cognitive impairment, which was associated with noninvasive markers of pulmonary vascular remodeling.

Summary: Peripheral neuropathy (PN) is a common complication in patients with prediabetes and Type 2 diabetes (T2D) and is often associated with dyslipidemia and mitochondrial dysfunction. Studies on a high-fat diet mouse model of prediabetes showed that dyslipidemia led to reduced mitochondrial membrane potential and impaired nerve impulse conduction. These results suggest that mitochondrial dysfunction may contribute to the pathogenesis of PN in prediabetes.

Summary: Proper growth and patterning of blood vessels are critical for embryogenesis. Chemicals or environmental hormones may interfere with vascular growth and cause developmental defects. In this study, a novel nitrobenzoate-derived compound, X8, was tested for its effects on vascular development using zebrafish as a model organism. The results showed that X8 treatment impaired the growth of blood vessels and caused circulatory defects in zebrafish embryos. Furthermore, X8 inhibited the expression of vascular genes and disrupted angiogenesis induced by exogenous stimuli. These findings suggest that X8 could be a potential antiangiogenic agent.

In two rat models of salt-dependent hypertension, the intrinsic and synaptic excitation of MNCVP caused by hypertonicity was differentially potentiated. Dietary salt increases the excitability of MNCVP through effects on the cell-autonomous and synaptic osmoresponsiveness of MNCVP.
