MICROSATELLITE MARKER DEVELOPMENT FOR THE FEDERALLY LISTED PRUNUS GENICULATA (ROSACEAE)

Premise of the study: Prunus geniculata is a federally listed species endemic to the central Florida scrub. To investigate levels and geographic partitioning of genetic variation in this species, we developed microsatellite markers for P. geniculata and its sister species, P. maritima. Methods and Results: Following a protocol optimized in our laboratory, we designed 36 primer pairs, of which 8 amplified across both species and showed informative polymorphism. Product size varied from 163 to 370 bp, and the number of alleles ranged from 5 to 27. H(e) across all populations ranged from 0.604 to 0.9155. Conclusions: Eight loci were useful at the population level and will be used for conservation genetic and phylogeographic studies of Prunus geniculata and P. maritima. Furthermore, from five to all eight of these loci can also be amplified in several other North American Prunus species.

Summary: Obtaining high-quality genomic DNA from plants for next-generation sequencing can be challenging due to secondary compounds within plant tissues. Researchers often need to test different extraction methods tailored to their specific species of interest. This study presents a rapid DNA extraction protocol using CTAB and sodium chloride, along with silica maxi-column cleanup, which is easy to implement, cost-effective, and scalable for diverse plant species.

Tragopogon dubius: Multiple introductions to North America and the formation of the New World tetraploids

Summary: Tragopogon is an important model for the study of whole-genome duplication, and researchers have found that introduced T. dubius plants from Europe have formed multiple tetraploid species in North America,with repeated formation involving only one of the ITS subtypes. The study also suggests that many entities traditionally considered as T. dubius should be recognized as distinct species.

Tragopogon pratensis: Multiple introductions to North America, circumscription, and the formation of the allotetraploid T. miscellus

Summary: Tragopogon (Asteraceae) is a genus that includes two recently formed allopolyploids, T. mirus and T. miscellus, in western North America. The introduction of diploid parents, T. dubius and T. porrifolius from Europe, played a role in the formation of these allopolyploids. In this study, we focused on the introduction of the second diploid parent, T. pratensis, into North America. Our findings show that T. pratensis as currently defined is polyphyletic, with at least four major ITS types in its native range. Two of these ITS types have been introduced from Europe into North America and are widespread in Canada and the U.S.A. The formation of T. miscellus in the western North America involved only one of these ITS types. Our research suggests that not all genotypes of T. pratensis may be capable of participating in the formation of allopolyploids. Additionally, we found that there are genetically distinct entities within T. pratensis that may be considered separate species. Further research involving phylogenetic analyses, taxonomy, morphology, and cytology are needed to fully understand the complexities of T. pratensis.
