Summary: This study investigated the effect of the APOE epsilon 2 allele on basal forebrain cholinergic system functional connectivity in cognitively normal and mild cognitive impairment patients. The results showed an interaction effect between APOE epsilon 2 genotype and functional connectivity, and the functional connectivity was associated with cognitive performance. Therefore, the APOE epsilon 2 genotype may have a protective role in mild cognitive impairment.

Summary: Studies have found that certain brain regions are activated during attentional deployment via long-term memories. We conducted an analysis of task-based functional connectivity to study the communication between these brain regions underlying long-term memory guided attention. Our results showed that different subnetworks, such as default mode, cognitive control, and dorsal attention, contribute differently to long-term memory guided attention, and the connectivity at the network level shifts based on attentional demands. We also found specific nodes within these subnetworks that play a crucial role in facilitating long-term memory guided attention.

Summary: The investigation of resting-state functional connectivity (rsFC) in asymptomatic individuals at genetic risk for Alzheimer's disease (AD) enables discovering the earliest brain alterations in preclinical stages of the disease. The APOE-epsilon 4 variant is the major genetic risk factor for AD, and previous studies have reported rsFC abnormalities in carriers of the epsilon 4 allele. Yet, no study has assessed APOE-epsilon 4 gene-dose effects on rsFC measures, and only a few studies included measures of cognitive performance to aid a clinical interpretation. Our data indicate that APOE-epsilon 4 shapes the functional architecture of the resting brain and favor the idea of a network-based functional compensation.

Summary: This study investigated tau deposition and its impact on functional connectivity in Alzheimer's disease through PET/MRI scans and neuropsychological assessments. Higher tau accumulation was found in specific brain regions of AD patients compared to healthy controls, leading to decreased functional connectivity. Patients with positive functional connectivity were older, had better cognitive performance, and lower tau accumulation compared to patients with negative functional connectivity. The combination of tau-PET and rs-fMRI could potentially help predict AD progression.
