Polymorphisms of APOE and LRP genes in Brazilian individuals with Alzheimer disease

Alzheimer disease (AD) is the most frequent cause of dementia in Western countries. Putative genetic risk factors for AD are polymorphisms in the apolipoprotein E (APOE) gene and in the low-density lipoprotein receptor-related protein (LRP) gene. Our objective was to investigate the role of the APOE coding region polymorphisms epsilon 2, epsilon 3, and epsilon 4 and APOE promoter variants A/T at position -491 and G/T at -219, as well as LRP polymorphism C/T, as risk factors for AD in Brazilian individuals. One hundred and twenty patients with probable AD, along with 120 controls were analyzed. A significant difference between patients and controls for 64 alleles was observed: frequency of this allele in AD was 0.31, and 0.10 in controls. Individuals with 2 FA alleles had a higher risk for AD than subjects with only 1 such allele; presence of 1 epsilon 2 allele proved protective. The presence of the T allele of the -219 polymorphism was also associated with an increased risk of AD, but this polymorphism is in linkage disequilibrium with APOE F polymorphisms. No significant differences between patients and controls were observed for -491 APOE or LRP polymorphisms. In this Brazilian population, both the epsilon 4 allele and T -219 polymorphism were associated with an increased risk for AD.

Summary: This study establishes a model of malignant astrocyte transformation by subjecting primary astrocytes from healthy adults to successive cycles of anchorage impediment. The transformed cells exhibit malignant characteristics and molecular analysis reveals the involvement of multiple signaling pathways and genes related to cell dedifferentiation. This model is significant for understanding glioblastoma treatment strategies and the underlying transformation mechanism.

Distribution of leptin receptors in the brain stem: possible route in the pathophysiology of neuromuscular control of airway resistance during sleep

Summary: This study aimed to investigate the distribution of leptin receptors in the brain stem and hypoglossal nucleus, and their association with sleep apnea. Immunohistochemical staining analysis was conducted on five cadavers, revealing the immunopositivity of leptin receptors in the motor neurons of the hypoglossal nucleus. These findings contribute to a better understanding of the neuromuscular control of airway collapse during sleep and the potential development of new drugs for improving upper airway tone in individuals with sleep apnea.

GBM Cells Exhibit Susceptibility to Metformin Treatment According to TLR4 Pathway Activation and Metabolic and Antioxidant Status

Summary: This study analyzed the effects of MET combined with TMZ treatment in two GBM cell lines, U87MG and A172, stimulated with LPS. MET treatment led to mitochondrial respiration blunting and oxidative stress in both cell lines, with U87MG cells exhibiting increased apoptosis after MET + LPS + TMZ treatment and A172 cells showing cell cycle arrest after MET + TMZ treatment. The observed differential response was associated with the metabolic status of the cells. This study highlights the potential of MET as a treatment option for GPM-GBM cases with an activated TLR4 pathway and suggests the use of combination therapies, such as an IL8 inhibitor for cases with upregulated CXCL8 and an antioxidant inhibitor for MTC-GBM cases.

Summary: This study aimed to investigate the key differences in discriminative networks and their main nodes between superagers and cognitively average elderly controls. The key networks that differentiated superagers and elderly controls were the default mode, salience, and language networks. Potential imaging biomarkers for predicting superagers can be provided by the functional connectivity in these networks. The 7T field holds promise for accurately detecting the functional connectivity patterns in superagers.

High performance older adults in a population-based sample with low education: Pieta study

Summary: This study investigated the presence of high-performance older adults (HPOA) in an oldest-old population with low education and explored associations with sociodemographic, clinical, and lifestyle variables. Results showed that 18 individuals fulfilled HPOA criteria, with an average age of 77.4 years and education level of 4.6 years. Younger age and lower scores on the Geriatric Depression Scale were associated with HPOA.

Summary: Diabetes mellitus is a risk factor for dementia and can induce neuronal dysfunctions through inflammation and other processes. This study found that obese individuals with diabetes had reduced GLUT4 expression and neuronal soma area in the hippocampus. Inflammatory activation decreased GLUT4 expression and negatively affected genes related to neuronal function. Inhibiting HDAC3 could potentially improve GLUT4 expression and counteract the negative effects of inflammation.

Machine Learning-Based Routine Laboratory Tests Predict One-Year Cognitive and Functional Decline in a Population Aged 75+ Years

Summary: This study used machine learning algorithms to assess the ability of routine laboratory variables in predicting cognitive and functional decline in older adults. The random forest model was found to be the most accurate in predicting cognitive decline, including variables such as triglycerides, glucose, and hemoglobin. For functional decline, platelet count, hemoglobin, and cortisol were identified as important features. The findings suggest that routine laboratory variables can be used to predict cognitive and functional decline in older populations using machine learning algorithms.

Current clinical and research practices on frontotemporal dementia in Brazil: a national survey

Summary: The aim of this study was to investigate the difficulties and limitations faced by Brazil in the diagnosis and management of frontotemporal dementia (FTD). A total of 256 Brazilian physicians participated in an online survey, and the results showed that the main difficulty in the diagnosis of FTD was the performance of genetic testing. Social cognition assessment and AD CSF biomarkers were considered useful for the diagnosis of FTD by 93% and 63% of participants, respectively.

Summary: The study investigates the value of using ACE-R combined with sociodemographic factors in the differential diagnosis between AD and bvFTD. The results show that combining ACE-R scores with age, sex, and education level can effectively differentiate AD from bvFTD.
