Newly appreciated roles for basophils in allergy and protective immunity

Basophils are evolutionarily conserved in many animal species, in spite of the fact that they account for <1% of peripheral blood leukocyte. This suggests that basophils have an indispensable and nonredundant role in vivo, even though they show some phenotypic similarity with tissue-resident mast cells. However, their functional significance remained uncertain long after Paul Ehrlich discovered them as blood-circulating cells with basophilic granules more than 130 years ago. The study of basophils has been far behind that of mast cells, owing to the rarity of basophils and the paucity of tools for their detection and functional analysis. Recent development of novel analytical tools, including basophil-depleting antibodies and genetically engineered mice deficient only in basophils, has greatly advanced basophil research and illuminated previously unrecognized roles of basophils. We now appreciate that basophils and mast cells play distinct roles in immune responses. Basophils have crucial roles in the development of acute and chronic allergic responses, the protective immunity against ecto- and endoparasites, and the regulation of acquired immunity, including the augmentation of humoral memory responses and the initiation of Th2 responses. Thus, basophils are no longer the neglected minority and are key players in the immune system.

Summary: E-cadherin expression in the hematopoietic system was studied using single-cell RNA-sequencing analyses and immunophenotyping. High levels of E-cadherin were found in progenitors within the granulocyte-monocyte progenitors (GMPs) fraction, with an enriched capacity to differentiate into basophils and mast cells. The transcriptional priming of these progenitors to the basophil and mast cell lineages was observed, with GATA-2 directly regulating E-cadherin expression in these lineages.

Basophil-derived IL-4 promotes cutaneous Staphylococcus aureus infection

Summary: Research has shown that mechanical injury to mouse skin through tape stripping predisposes the skin to superficial infection with S. aureus. This infection is promoted by basophil-derived IL-4, which inhibits the production of IL-17A and facilitates S. aureus infection. IL-4 acts on multiple checkpoints to suppress the cutaneous IL-17A response, but blocking the IL-4 receptor can enhance Il17a expression and bacterial clearance.

Role of Basophils in a Broad Spectrum of Disorders

Proinflammatory role of basophils in oxazolone-induced chronic intestinal inflammation

Summary: This study investigates the roles of basophils in human ulcerative colitis (UC) and oxazolone (OXA) colitis using genetically engineered mice. The results show that basophil infiltration is significantly increased in patients with UC and correlates with disease severity. Depletion of basophils improves inflammation in mouse models of colitis. These findings suggest that basophils play a proinflammatory role in chronic intestinal inflammation such as UC.

Summary: Tissue-specific mouse models are important tools for studying the role of different cell compartments and genes in diseases. A new knock-in mouse model is described here, allowing selective expression of fluorescent protein tdTomato and CRE recombinase in the basophil compartment. This model enabled the identification and study of basophils, and breeding with another mouse model resulted in basophil-deficient mice with no abnormalities in other cell compartments. The study showed that basophils play a nonredundant role in pristane-induced lupus-like disease, both in its induction and amplification.
