SIRT1 regulatess, sphingolipid metabolism and neural differentiation of mouse embryonic stem cells through c-Myc-SMPDL3B

Summary: The study reveals that sphingolipid metabolism is transcriptionally controlled by SIRT1 in mESCs, and deletion of SIRT1 leads to accumulation of sphingomyelin, affecting membrane fluidity and neural differentiation. This key regulatory mechanism may provide insights for potential treatment of human neurological diseases through pharmacological manipulation of SIRT1-mediated sphingomyelin degradation.

Metabolic and epigenetic regulation of endoderm differentiation

Yi Fang, Xiaoling Li

Summary: The endoderm, one of the three primary germ layers, gives rise to important organs, but growing cells/tissues from endodermal organs in vitro is challenging. Human pluripotent stem cells (hPSCs) hold great promise for generating endodermal cells for therapeutic development. Recent research has revealed the active role of metabolism and epigenetics in regulating endoderm differentiation.

Uterine-specific SIRT1 deficiency confers premature uterine aging and impairs invasion and spacing of blastocyst, and stromal cell decidualization, in mice

Summary: A distinct age-related alteration in the uterine environment has been identified as a prevalent cause of reproductive decline in older female mice. Sirtuin 1 (SIRT1), a multifunctional enzyme involved in regulating cell viability, senescence, and inflammation during aging, is reduced in aged decidua. Uterine-specific ablation of Sirt1 gene accelerates premature uterine aging and impairs invasion and spacing of blastocysts, as well as decidualization of stromal cells.
