Can cocaine use be evaluated through analysis of wastewater? A nation-wide approach conducted in Belgium

Aims Cocaine is the second most-used illicit drug world-wide and its consumption is increasing significantly, especially in western Europe. Until now, the annual prevalence has been estimated indirectly by means of interviews. A recently introduced and direct nation-wide approach based on measurements of the major urinary excreted metabolite of cocaine, benzoylecgonine, in wastewater is proposed. Design Wastewater samples from 41 wastewater treatment plants (WWTPs) in Belgium, covering approximately 3 700 000 residents, were collected. Each WWTP was sampled on Wednesdays and Sundays during two sampling campaigns in 2007-08. Samples were analysed for cocaine (COC) and its metabolites, benzoylecgonine ( BE) and ecgonine methylester (EME) by a validated procedure based on liquid chromatography coupled with tandem mass spectrometry. Concentrations of BE were used to calculate cocaine consumption (g/day per 1000 inhabitants) for each WWTP region and for both sampling campaigns (g/year per 1000 inhabitants). Findings Weekend days showed significantly higher cocaine consumption compared with weekdays. The highest cocaine consumption was observed for WWTPs receiving wastewater from large cities, such as Antwerp, Brussels and Charleroi. Results were extrapolated for the total Belgian population and an estimation of a yearly prevalence of cocaine use was made based on various assumptions. An amount of 1.88 tonnes (t) per year [standard error (SE)0.05 t] cocaine is consumed in Belgium, corresponding to a yearly prevalence of 0.80% (SE 0.02%) for the Belgian population aged 15-64 years. This result is in agreement with an earlier reported estimate of the Belgian prevalence of cocaine use conducted through socio-epidemiological studies (0.9% for people aged 15-64 years). Conclusions Wastewater analysis is a promising tool to evaluate cocaine consumption at both local and national scale. This rapid and direct estimation of the prevalence of cocaine use in Belgium corresponds with socioepidemiological data. However, the strategy needs to be refined further to allow a more exact calculation of cocaine consumption from concentrations of BE in wastewater.

Gestational Exposure to Plastic Additives and Associations with Placental Function-Related Genes

Summary: This study aimed to characterize gestational exposure to plasticizers and antioxidants, and explore the associations with placental function related genes. Several chemicals showed significantly different concentrations between preterm and full-term deliveries, and were associated with mRNA expression levels of selected genes. The associations exhibited chemical-specific patterns and significantly differed between preterm and full-term deliveries.

Metabolic signature of HepaRG cells exposed to ethanol and tumor necrosis factor alpha to study alcoholic steatohepatitis by LC-MS-based untargeted metabolomics

Summary: Despite being common, alcoholic liver disease remains poorly understood, especially in its early stages. In this study, alcoholic steatohepatitis was simulated in vitro to investigate the metabolic alterations and identify affected pathways. The researchers discovered key findings, such as the influence of TNF-alpha on hepatic triglycerides and the detection of FAHFA-containing triglycerides in human hepatocytes, which may play an important role in the progression of alcoholic steatohepatitis. Additionally, ethoxylated phosphorylcholine was identified as a potential new biomarker of ethanol exposure.

Assessment of silicone wristbands for monitoring personal exposure to chlorinated paraffins (C8-36): A pilot study

Investigating the Potential of Drift Tube Ion Mobility for the Analysis of Oxidized Lipids

Summary: Epilipids, a subset of the lipidome, play a crucial role in regulating redox lipid metabolism and influencing cell fate decisions. Ion mobility spectrometry (IMS) combined with liquid chromatography-high-resolution mass spectrometry (LC-HRMS) can improve the peak capacity and separation ability for the analysis of oxidized complex lipids. This study demonstrates the advantages and limitations of implementing IMS in LC-HRMS workflows and provides insights into the annotation of oxidized complex lipids.

Identification and characterization of quaternary ammonium compounds in Flemish indoor dust by ion-mobility high-resolution mass spectrometry

Summary: Quaternary ammonium compounds (QACs) in European indoor dust were identified, characterized, and semi-quantified using ion-mobility high-resolution mass spectrometry. Targeted QACs (21) and suspect QACs (17) were detected with high detection frequencies. The concentrations of individual QACs were estimated and daily intakes for adults and toddlers were calculated. This study provides important insights into the potential human exposure to QACs.

An international investigation of chlorinated paraffin concentrations and homologue distributions in indoor dust*

Summary: In this study, dust samples from Japan, Australia, Colombia, and Thailand were analyzed for different chain lengths of chlorinated paraffins. The results showed that chlorinated paraffins were detected in all samples, with the highest concentrations found in samples from Colombia and Thailand. Based on current toxicological data, it was determined that short-chain and medium-chain chlorinated paraffins in indoor dust do not pose a health risk through ingestion.

A retrospective investigation of feather corticosterone in a highly contaminated white-tailed eagle (Haliaeetus albicilla) population

Summary: English Summary: Exposure to persistent organic pollutants (POPs), such as organochlorines (OCs) and polybrominated diphenyl ethers (PBDEs), can have adverse health effects on wildlife. White-tailed eagles in the Baltic ecosystem have suffered population declines due to severe exposure to DDT and PCB. In this study, feathers from breeding WTE pairs were analyzed to investigate the annual variations in feather corticosterone (fCORT), POPs, and dietary proxies. The results did not support fCORT as a relevant biomarker of contaminant-mediated effects in WTEs, but fCORT can be used as a non-destructive assessment of long-term stress physiology in wild raptors.

Hair as an alternative matrix to assess exposure of premature neonates to phthalate and alternative plasticizers in the neonatal intensive care unit

Summary: This study reports metabolites of phthalate and alternative plasticizers in neonatal hair samples as biomarkers for exposure to these plasticizers. Results showed that hair sampled from premature neonates after a NICU stay contained significantly higher metabolite concentrations of both phthalates and alternative plasticizers compared to healthy control neonates. Prolonged NICU exposure to respiratory support devices and gastric tubes was correlated with increased concentrations in hair samples.

Summary: This study evaluated the exposure levels of children and women in Europe over time. The results showed that exposure levels to chemicals restricted at EU level visually decreased, while the levels for some of their substitutes increased. These findings are important for policy efficacy monitoring and highlight the role of HBM in supporting policies.

Summary: The knowledge about the effects of organophosphate flame retardants on children's neurodevelopment is limited. This research aimed to evaluate the association between exposure to these flame retardants and children's neurodevelopment. The study found a small, negative trend between neurodevelopment and exposure to BDCIPP and DPHP, but the evidence is limited. Further investigations are needed to explore the timing of exposure and the effect of other metabolites.
