Summary: Pain can be induced by tissue injuries, diseases, and infections. The interactions between the peripheral nervous system (PNS) and immune system play crucial roles in pain sensitization processes. Immune cells release mediators that activate nociceptors and promote sensitization, while also contributing to PNS repair and pain resolution.

Summary: With the increasing prevalence of neurological disorders in an aging population, there is a critical need to better understand exogenous strategies for nervous system repair. Stem cell therapy offers a promising approach, but is limited by various factors. Researchers propose the use of autologous cells derived from the peripheral nervous system as an alternative source for CNS repair, which has shown neurogenic potential and eliminates concerns around tumorigenic risk, ethical considerations, and immune-mediated rejection.

Summary: The study revealed that the absence of NCS1 in the brain leads to reduced dendritic complexity and spine density, which may be associated with deficits in memory acquisition. RNA sequencing results showed that NCS1 modulates gene expression related to neuronal morphology and development.

Summary: The peripheral nervous system plays a crucial role in regulating immune responses through sensory and effector circuits. Neurons can directly respond to inflammatory signals and advancements in single-cell sequencing have allowed for a better understanding of distinct neuronal subsets. This neuro-immune interaction may lead to novel therapeutic approaches for a variety of diseases, including chronic inflammation.

Summary: This study finds that IM33 and SLPI are upregulated in old flies and old mice, respectively, suggesting their key roles in aging. Knockdown of IM33 in glia leads to increased gut ROS levels and altered gut microbiota composition, resulting in a shortened lifespan. Dysbiosis also induces sleep fragmentation through the activation of insulin-producing cells in the brain. Identifying molecular mediators of these processes could contribute to the development of innovative strategies for extending lifespan.

Summary: Research has shown that in chronic demyelinating diseases, increased neuronal cholesterol synthesis plays a critical role in remyelination, especially when lipid recycling is defective in the affected area. This finding contributes to understanding disease progression and has implications for therapeutic strategies in patients with multiple sclerosis (MS).

Summary: The peripheral nervous system is a heterogeneous system with diverse functions, including communication between the brain and the body, control of development and regeneration. It can be divided into sensory, motor, autonomic and enteric domains. Recent advances in single-cell transcriptomics have revealed new neuronal subtypes and improved cell-type classifications. This article discusses the roles, origin and heterogeneity of the peripheral nervous system, highlighting recent discoveries and future perspectives.

Summary: The study found that nucleoredoxin (NXN) plays an important role in regulating thermal sensation and nociception. The loss of NXN in neurons leads to increased sensitivity to cold temperature and heightened heat nociception in mice. These abnormal responses are associated with changes in calcium influx, mitochondrial dysfunction, and increased density of neurite trees in sensory neurons.

Summary: This study explores the genetic expression patterns during the development of peripheral nervous system (PNS) in invertebrate chordates, and compares them with vertebrates. The results suggest that the ventral PNS in invertebrates may not be homologous with each other, but there is a high homology between ventral PNS and dorsally forming PNS in vertebrates.

Summary: The study found that higher parasympathetic nervous system activity, as indicated by higher RSA, was associated with increased expression of Type I interferon response genes in circulating leukocytes, but not with inflammatory gene expression. This suggests a previously unrecognized immunoregulatory aspect of autonomic nervous system function and highlights a potential biological pathway by which parasympathetic nervous system activity may impact health.

Summary: While individual neurons are the building blocks of the nervous system, they function by collaborating in neuronal circuits with specific patterns of synaptic connectivity. Understanding how these circuits assemble and evolve can help bridge the gap between individual neuron biology and brain function, leading to insights into neural computations and behavior.

Summary: This study investigated the role of spinal neuronal GRK2 in chemotherapy-induced peripheral neuropathy (CIPN) and the potential preventive effect of electroacupuncture (EA). The findings suggest that neuronal GRK2 may be a potential target for CIPN intervention and EA treatment can mimic the effects of neuronal GRK2 to alleviate pain and sensory deficits.
