Retinal function after intravitreal injection of autologous bone marrow-derived mesenchymal stromal cells in advanced glaucoma

Carolina Assis P. Vilela, Andre Messias, Rodrigo T. Calado, Rubens C. Siqueira, Marcelo Jordao L. Silva, Dimas T. Covas, Jayter S. Paula

Summary: The study reported on the use of bone marrow-derived mesenchymal stem cells (MSCs) in treating advanced glaucoma, with no improvement seen in visual acuity or visual field post-treatment. Although one patient experienced retinal detachment and proliferative vitreoretinopathy after 15 days, the other patient showed stable ERG responses throughout the study period. Further research involving modified MSCs is needed to address the lack of visual function improvement observed in the study.

SCORE2 Report 17: Macular thickness fluctuations in anti-VEGF-treated patients with central or hemiretinal vein occlusion

Ingrid U. Scott, Neal L. Oden, Paul C. VanVeldhuisen, Michael S. Ip, Barbara A. Blodi

Summary: Greater fluctuations in central subfield thickness as assessed by standard deviation and Zigzag measures were negatively associated with Month 12 visual acuity letter score. However, early post-treatment visual acuity letter score is a stronger predictor of visual acuity outcomes than the central subfield thickness fluctuation measures.

Summary: This study provides a comprehensive overview of the CNGA3 variant spectrum in 1060 genetically confirmed ACHM patients, with 385 of them carrying likely disease-causing variants in CNGA3. By combining their own genetic data with literature and public database records, the researchers extended the CNGA3 variant spectrum to a total of 316 variants, with 244 of them interpreted as likely disease-causing. They also reported 48 novel likely disease-causing variants, with 24 of them being missense substitutions. Extensive in silico analyses and functional data were provided to advance the pathogenicity assessment of the identified variants.

Baseline Characteristics and Outcomes After Anti-Vascular Endothelial Growth Factor Therapy for Macular Edema in Participants With Hemiretinal Vein Occlusion Compared With Participants With Central Retinal Vein Occlusion Study of Comparative Treatments for Retinal Vein Occlusion 2 (SCORE2) Report 18

Ingrid U. Scott, Neal L. Oden, Paul C. VanVeldhuisen, Michael S. Ip, Barbara A. Blodi

Summary: This study compared the baseline characteristics, treatment burden, and outcomes of macular edema in HRVO and CRVO patients. The study found a higher proportion of Black participants in HRVO group compared to CRVO group, and no significant differences in the frequency of macular edema treatments between the two groups. Although CRVO eyes had worse visual acuity and more macular edema compared to HRVO eyes at baseline, the magnitude of VALS improvement, central retinal thickness response to anti-VEGF therapy, and treatment burden were similar between the groups.

Month 60 Outcomes After Treatment Initiation With Anti-Vascular Endothelial Growth Factor Therapy for Macular Edema Due to Central Retinal or Hemiretinal Vein Occlusion

Ingrid U. Scott, Paul C. Vanveldhuisen, Neal L. Oden, Michael S. Ip, Barbara A. Blodi

Summary: This study investigates the long-term outcomes of using aflibercept or bevacizumab to treat macular edema. The results show that while there is significant improvement in visual acuity and central subfield thickness during the initial treatment period, the improvement lessens when treatment is at the discretion of the investigator. However, most patients continue to receive treatment in the 5-year period.

SCORE2 Report 20: Relationship of Treatment Discontinuation With Visual Acuity and Central Subfield Thickness Outcomes

Ingrid U. Scott, Neal L. Oden, Paul C. Vanveldhuisen, Michael S. Ip, Barbara A. Blodi

Summary: This study aimed to investigate the relationship between early discontinuation of anti-vascular endothelial growth factor (anti-VEGF) treatment and baseline factors and outcomes. The study found that compared to the continuous treatment group, patients who discontinued treatment early were younger and had a higher proportion of black individuals. They also had a higher proportion of complete resolution of macular edema and a lower central thickness over follow-up, but their visual acuity was not better than the continuously treated and intermittently treated groups. The results support the need for continued monitoring and individualized treatment for patients with macular edema from central or hemiretinal vein occlusion.

Summary: This study aims to quantify the association between visit adherence and visual acuity in patients with retinal vein occlusions. Visit adherence was measured by the number of missed visits, average and longest visit interval, and average and longest missed days. The study found that the number of missed visits was associated with a decrease in visual acuity, but the length of visit interval was not associated with changes in visual acuity.

Nonlinear Relationship of Retinal Thickness and Visual Acuity in Central Retinal and Hemiretinal Vein Occlusion

Ingrid U. Scott, Neal L. Oden, Paul C. Vanveldhuisen, S. Michael, B. A. Blodi

Summary: There is a nonlinear association between central subfield thickness (CST) on spectral-domain OCT and concurrent visual acuity letter score (VALS) in eyes treated initially with afli-bercept or bevacizumab for macular edema associated with central retinal vein occlusion (CRVO) or hemiretinal vein occlusion (HRVO). The best expected VALS is observed when post-treatment CST is close to the estimated inflection points.

Combined intravitreal dexamethasone and bevacizumab injection for the treatment of persistent diabetic macular edema (DexaBe study): a phase I clinical study

Francyne Veiga Reis, Pedro Dalgalarrondo, Jose Edisio da Silva Tavares Neto, Murilo Wendeborn Rodrigues, Ingrid U. Scott, Rodrigo Jorge

Summary: This study aimed to investigate the safety and effects of combined intravitreal injection of dexamethasone and bevacizumab in patients with refractory diabetic macular edema (DME). The results indicated that the combined therapy led to increased intraocular pressure and the need for anti-glaucomatous eye drops, as well as reduced central sub-foveal thickness. However, there was no significant improvement in best-corrected visual acuity.

OCT Grading System of Macular Infarction Predicts Vision in Participants With Central Retinal or Hemiretinal Vein Occlusion: A Secondary Analysis of SCORE2

A. U. Adrian, I. P. Michael, Barbara A. Blodi, Ingrid U. Scott, Neal L. Oden, Paul C. van Veldhuisen, David Sarraf

Summary: This study found a strong correlation between the severity of macular infarction measured by hyper-reflectivity of the middle and inner retinal layers and long-term visual outcomes in participants with central retinal vein occlusion. Despite significant improvement in macular edema after treatment, participants with more severe macular infarction showed worse visual outcomes, suggesting that macular infarction may play a role in visual acuity even after anti-VEGF treatment.
