Summary: The study found that 14α-methylsterols replace ergosterol to allow Candida albicans to survive Fluconazole (FLC), but Erg251 inactivated by CZ66 results in the loss of 14α-methylsterol synthesis and cell death of Candida albicans treated with FLC. Establishing Erg251 as a synergistic lethal target protein of FLC should direct research to identify specific small molecule inhibitors of 14α-methylsterol synthesis and open the way to abolishing fungal FLC tolerance.

Summary: This study evaluated the potential of successive applications of sub-lethal doses of antimicrobial photodynamic therapy (aPDT) mediated by Photodithazine (PDZ) and curcumin (CUR) associated with LED in the viability, reactive oxygen species (ROS) production, and gene expression of Candida albicans. Results showed that CUR-mediated aPDT could induce C. albicans resistance in biofilms, while PDZ-mediated aPDT had a susceptible effect on C. albicans cells in biofilms. The production of ROS was higher in the biofilms than in the planktonic cultures, and SOD1 expression was the highest in PDZ-mediated aPDT.
