Herpes simplex virus type 2 infection is a quite common but frequently asymptomatic, therefore undiagnosed condition. Genital HSV-2 infection may cause neonatal herpes, enhances HIV transmission and may play a role in infertility. To evaluate the prevalence of HSV-2 in Hungary we tested 2500 serum samples for the presence of anti-HSV-2 IgG by ELISA method. According to our results Hungary belongs to the low-infected countries, the HSV-2 seroprevalence grows with age and is significantly higher among women than in men. We also examined the serostatus of 512 pregnant women and 539 women attending infertility clinics. Results show that the HSV-2 prevalence is significantly higher among women attending infertility clinics and the seropositivity of pregnant women is similar to that of the general Hungarian women population with the same age.

Summary: In this study, a genome-wide association analysis for resting QT interval identified 40 loci, including 13 novel ones, explaining approximately 11% of the trait variance. Candidate genes at the novel loci were found to be involved in myocardial structure and arrhythmogenic cardiomyopathy, providing new insights into the mechanisms underlying abnormal myocardial repolarization and cardiovascular mortality.

Frequency and management of medically actionable incidental findings from genome and exome sequencing data: a systematic review

Amal Elfatih, Idris Mohammed, Doua Abdelrahman, Borbala Mifsud

Summary: The application of whole genome/exome sequencing technologies in clinical genetics and research has led to the discovery of incidental findings unrelated to the primary purpose of genetic testing. While guidelines for reporting pathogenic variants with medical actionability have been published, consensus guidelines for variant reporting and classification are still lacking. A systematic literature review was conducted to understand the variable reporting and classification methods for medically actionable variants across different populations, highlighting areas that require further consideration or adjustment.

Structure-Based Virtual Screening and Functional Validation of Potential Hit Molecules Targeting the SARS-CoV-2 Main Protease

Summary: The COVID-19 pandemic caused by the coronavirus disease 2019 (COVID-19) has had a significant impact on lives and economies worldwide. Vaccines have been developed, but their effectiveness has been variable due to challenges in production, logistics, distribution, and vaccine hesitancy. Additionally, existing vaccines are less effective against new variants of the SARS-CoV-2 virus and immunity from vaccination diminishes over time. This highlights the need for improved treatment options, such as inhibiting the SARS-CoV-2 main protease (M-pro). Drug repurposing using virtual screening has identified potential inhibitors from natural products and FDA-approved drugs. Experimental validation has shown promising results, suggesting that existing drugs and natural products could be explored for the treatment of COVID-19.

Summary: Identification of novel BRCA1 variants outpaces their clinical annotation, emphasizing the need for accurate computational methods for risk assessment. In this study, a BRCA1-specific machine learning model was developed to predict the pathogenicity of BRCA variants, and its performance was evaluated on different sets of variants. The model showed high accuracy in predicting pathogenicity and functional consequence, and identified potentially pathogenic variants in unreviewed BRCA1 variants.
