Summary: The formation of tertiary lymphoid organs (TLOs) in the mesentery is a feature of Crohn's disease, blocking cellular and molecular outflow from the gut and causing lymph leakage and backflow. TNF neutralization at early stages can restore lymph transport, but developed chronic TLOs resist regression and restoration of flow after TNF neutralization. TNF stimulation of lymphatic endothelial cells disrupts immune cell transport and valve-associated gene induction, contributing to Crohn's disease pathology.

Summary: The lymphatic vasculature plays a crucial role in maintaining tissue homeostasis and immune surveillance in the lung. Changes in the lung lymphatic system have been observed in various respiratory diseases, and recent research suggests that lymphatic dysfunction may contribute to the development and progression of lung disease. Further understanding of the mechanisms underlying these changes could lead to new therapeutic strategies.

Summary: The study used single-cell RNA sequencing to investigate transcriptional changes in lymphatic endothelial cells in lymph nodes draining naive and inflamed skin. It was found that subsets of these cells responded individually to skin inflammation, suggesting distinct functions under pathological conditions. Dysregulated genes during inflammation were identified, including an up-regulation of CD200, potentially involved in immune regulation. Additionally, in silico analysis predicted interactions of these cells with diverse immune cells in the lymph nodes, showing similarities in transcriptional changes in different skin inflammation settings.

Summary: The study shows that mannose supplementation can alleviate symptoms of inflammatory bowel disease, including colitis. It improves intestinal barrier integrity, reduces the release of proteins, and prevents mitochondrial dysfunction. Mannose also synergizes with the conventional drug mesalamine in colitis treatment.

Summary: This study presented the experience of lymphatic transfer in 30 patients with extremity lymphedema and 15 patients with post-traumatic lower extremity soft tissue defects. Results indicated that LFT may be a valid treatment option, reducing lymphedema index and improving stability in patients.

Summary: The study revealed that in the process of cardiac hypertrophy and failure, there is remodeling of lymphatic vessels and reduced lymphatic efficiency; the number of lymphatic endothelial cells in failing hearts is positively correlated with cardiac function and a subset of cardiac macrophages; macrophages identified by LYVE-1 play a key role in promoting lymphangiogenesis.

Summary: Intestinal inflammation is the underlying cause of colitis and inflammatory bowel diseases, influenced by genetic and environmental factors. Alteration in neuraminidase activity may trigger disease onset, and inhibition of Neu3 can protect against severe colitis development.

Summary: Studies have shown that impairment in lymphatic transport is linked to the onset and progression of atherosclerosis. The downregulation of low-density-lipoprotein receptor (LDLR) expression is involved in lymphatic dysfunction associated with early atherosclerosis. Enhancing lymphatic function with VEGF-C 152s can delay the formation of atherosclerotic plaques. The mechanisms by which LDLR protects against lymphatic function impairment are unclear, but it may affect chromosome duplication and VEGFR-3 expression.

Summary: Inactivation of Foxc2 in adult lymphatic endothelium leads to a stepwise intestine-to-lung systemic failure, compromising gut epithelial barrier and promoting dysbiosis. The loss of Foxc2 skewed the specialization of lymphatic endothelial subsets, resulting in populations with mixed identities and emergence of lymph node-like endothelial cells.
